ObjectiveTo investigate the expression of basic fibroblast growth factor (bFGF) and transformation-specific-l(Ets-1) in normal cervical tissue, cervical intra-epithelial neoplasia (CIN) and cervical cancer, analyze their relationship with the clinical pathological features. and reveal the correlation between the two factors,explore their effects in cervical carcinoma angiogenesis, development, invasion and metastasis.MethodImmunohistochemical stain (IHC) was used to investigate the expression of bFGF and Ets-1 in 30 cases of normal cervical tissue,60 cases of cervical intraepithelial neoplasia and 62 cases of cervical carcinoma tissue.and observe the clinical stage, histological grade, tumor size, lymph node metastasis.Adopt SPSS18.0 was used for statistically analysis, comparing the difference and correlation of target factors in those three different cervical lesion tissues.Resultsâ‘ IHC shows that bFGF is localized in the cytoplasm of cancer cells with yellow or brown colour in different tissues of cervical lesion. The positive expression rate of bFGF in normal cervical tissue, cervical intraepithelial neoplasia and cervical carcinoma is 10.00%,63.33%,90.32% respectively. The difference of bFGF expression in the three groups is statistical significance (P<0.01), comparison between two groups, normal uterine cervix and cervical cancer group, the difference was statistically significant (P<0.01), cervical intraepithelial neoplasia (CIN) group and the cervical cancer group, the difference was statistically significant (P<0.01), normal uterine cervix and cervical intraepithelial neoplasia (CIN) group, the difference was statistically significant (P<0.01).â‘¡The positive rates of bFGF in cervical intraepithelial neoplasia (CINI, CINâ…¡,CINâ…¢) is 35.00%,75.00%, and 80.00%, compared among the three groups, the difference was significant (P<0.05). Comparison between groups: CINâ… compared with CINâ…¡(P<0.05), statistically significant difference; CINâ… compared with CINâ…¢(P<0.01), the difference was statistically significant; CINII compared with CINIII (P>0.05), the difference was not statistically significant.â‘¢bFGF expression is closely rated clinical stage of cervical cancer, differentiation, tumor size, lymph node metastasis, expression was statistically significant (P<0.05). The later in the clinical stage, tumor differentiation, the lower the larger the lymph node metastasis of carcinoma of the higher, but not with patient age, pathological types.â‘£Ets-1 expression is yellow or brownish yellow fine granular distributing in cancer cells,endothelial cells, and interstitial infiltration of lymphocytes positive. Ets-1 in normal cervical tissue, cervical intraepithelial neoplasia (CIN), cervical cancer tissues were 36.66%,43.33%,77.41%, compared among the three groups difference was statistically significant (P<0.01).compared between two groups, normal uterine cervix and cervical cancer group, the difference was statistically significant (P<0.01), cervical intraepithelial neoplasia (CIN) and cervical group cancer group, the difference was statistically significant (P <0.01), Normal uterine cervix and cervical intraepithelial neoplasia (CIN) group, no significant difference (P>0.05).⑤The positive rates of Ets-1 in cervical intraepithelial neoplasia (CINâ… , CINâ…¡,CINâ…¢) is 15.00%,55.00%,60.00%, compared among the three groups difference was statistically significant (P<0.01). Between the two groups:CINâ… compared with CINâ…¢, P<0.01, the difference was statistically significant; CINâ… compared with CINâ…¡, P<0.01, the difference was statistically significant; CINâ…¡and CINâ…¢comparison, P>0.05, no statistical difference significance.â‘¥The expression of Ets-1 has closely relation to Clinical stage of cervical cancer, Differentiation, tumor size, lymph node metastasis, (P<0.05), in the later clinical stage, lower differentiation degree, tumor size, there are lymph node metastasis in carcinoma of the higher, but not with patient age, pathological types, (P>0.05).⑦bFGF and Ets-1 expression in cervical cancer are closely linked, moderate correlation is positive (Spearman correlation coefficient rs=0.607). â‘ from normal cervical tissueâ†'CINâ†'cervical cancer, The expressions of bFGF and Ets-1 protein tended to increase along with pathogenetic condition progression, suggested that bFGF and Ets-1 possibly played an important role in the genesis of cervical cancer.â‘¡The expressions of bFGF and Ets-1 protein had no statistical relation with age, which suggested that bFGF and Ets-1 were not major fators to promoting the occurrence of more young women with cervical cancer.â‘¢The expressions of bFGF and Ets-1 had significant relationship with clinical stage, tumor size and lymphatic metastasis, which suggested that Ets-1 may play an important role in the development, invasion and metastasis of cervical cancer together with bFGF.â‘£with cervical intraepithelial neoplasia grade increased, the positive rate of bFGF and Ets-1 protein also increased, suggesting that bFGF and Ets-1 in cervical lesions may play an important role in the development. Both can also assist the clinical differentiation is difficult to identify the CINâ… and CINâ…¡, for treatment after a reasonable basis to prevent over-treatment and missed treatment opportunity.⑤The expressions of bFGF and Ets-1 protein expression in cervical cancer was positively correlated, indicating that bFGF and Ets-1 in cervical cancer to develop in the course of the synergistic effects. bFGF and Ets-1 can be considered as the degree of malignancy of cervical cancer, cervical cancer invasion and metastasis of clinical prediction reference two important and valuable therapeutic targets, which in early cervical cancer diagnosis and prognosis of patients with cervical cancer may have important reference value.
