| [Objective]To study the expression and significance of EGFL7 and PTEN in human glioma tissues,in order to evaluate influences of the expression and significance on neovascularization and FAK-phosphorylation.[Methods]The expression of EGFL7 and PTEN in 56 cases of human glioma and 8 cases of normal brain tissues were detected by immunohistochemistry test (SP method) and their correlations of clinicopathologic features were analyzed, MVD and FAKpY397 were detected also.[Results]1,The expression of EGFL7 protein by human Gliomas was dctected, EGFL7 protein located in plasm of tumor cells and endothelial cells; There was a significant difference about the positive rates of EGFL7 between normal brain tissue and gliomas(P<0.01), immunohistochemistry showed that the positive rate of EGFL7 in gliomas and normal brain tissue was 75.0%(42/56) and 0%(0/8) respectively;With the pathological grade of tumors being higher,the higher expression level of EGFL7 showed(P<0.01);The expression levels of EGFL7 protein in gliomas were not correlated with sex ,age and smoking(P>0.05).2,The expression of PTEN protein was dctected in both human Gliomas and normal brain tissue,PTEN protein located in plasm of tumor cells and normal brain cells;There was a significant difference about the positive rates of PTEN between normal brain tissue and gliomas(P<0.01), immunohistochemistry showed that the positive rate of PTEN in gliomas and normal brain tissue was about 48.2%(23/56) and 100 % (8/8) respectively;With the pathological grade of tumors being higher ,the lower expression level of PTEN showed(P<0.01);The expression levels of PTEN protein in gliomas were not correlated with sex ,age and smoking(P>0.05).3,There was a significant difference about the positive rates of FAKpY397 between normal brain tissue and gliomas(P<0.05), immunohistochemistry showed that the positive rate of FAKpY397 in gliomas and normal brain tissue was about 73.2%(41/56) and 12.5 % (1/8) respectively;With the pathological grade of tumors being higher ,the higher expression level of FAKpY397 showed(P<0.01).4,There was a significant difference on MVD between normal brain tissue and gliomas(P<0.01),With the pathological grade of gliomas being higher ,the higher level of MVD showed(P<0.01).5,There was positive correlation between EGFL7 protein and FAKpY397 expression in gliomas(r=0.314, P<0.01),and EGFL7 protein and MVD showed the same results in gliomas(t= 26.55,P<0.01);There was negative correlation between PTEN protein and FAKpY397 expression in gliomas(r=-0.284, P<0.01),and PTEN protein and MVD showed the same results in gliomas(t=-17.414,P<0.01); There was negative correlation between EGFL7 protein and PTEN protein in gliomas(r=-0.382, P<0.01).[Conclusions]1,The expression of EGFL7 protein was dctected both in tumor cells and endothelial cells of human Gliomas,the level of EGFL7 expression direetly correlates with the degree of malignancy,MVD and FAKpY397 of human gliomas and may contribute to their rapid growth and invasion. Our results indieate that EGFL7 may be a potential indicator for evaluating glioma invasion.2,The expression of PTEN are significantly correlated to the tumor grade of gliomas, showing that absent expression of PTEN might act one importance on glioma occurrence and invasion.PTEN protein may be a potential indicator for predicting glioma invasion. The negative correlation between PTEN protein and FAKpY397 or MVD showed the control of PTEN to glioma invasion may correlat with FAK-phosphorylation, indicated PTEN might also act one importance on glioma neovascularization.3,The negative correlation between EGFL7 protein and PTEN protein in gliomas revealed one potential connection,awaiting more thorough research. EGFL7 and PTEN were potential indicator for evaluating glioma invasion,might become a prognostic marker for appreciating prognosis of the degree of malignancy in the future. |
