Objective:To investigate the expression of claudin-1 and claudin-3 in the eutopic and ectopic of women with endometriosis and to evaluate the role of claudin-1 and claudin-3 on the pathogenesis of endometriosis; To investigate the expression of claudin-1 and claudin-3 in the peritoneum and to evaluate the dependability between peritoneum and endometriosis.Methods:Forty women with endometriosis and thirty controls were studied. The expression levels of claudin-1,claudin-3 in eutopic and ectopic endometrium of women with endometriosis and normal endometrium of control group were measured by immunohistochemistry. Also the expression levels of claudin-1 and claudin-3 in normal peritoneum and peritoneum including ectopic focus of women with endometriosis and normal peritoneum of control group were measured by immunohistochemistry.Results:Compared with those of controls and eutopic endometrium, the expression of claudin-1,claudin-3 was found to be significantly decreased in ectopic endometrium of women with endometriosis(P<0.01). Compared with those of controls, the expression of claudin-1,claudin-3 was not found to be significantly dereased in eutopic endometrium of women with endometriosis(P>0.05). Expression of claudin-1 has no significant difference between normal peritoneum and peritoneum including ectopic focus of women with endometriosis and normal peritoneum of control group(P>0.05). Expression of claudin-3 also has no significant difference between normal peritoneum and peritoneum including ectopic focus of women with endometriosis and normal peritoneum of control group(P>0.05). No correlation was discovered between claudin1,3 and Staging of endometriosis (P>0.05)Conclusions:claudin-1,claudin-3 might be correlated with development of endometriosis due to its significant down regulation expression in ectopic endometrium of women with endometriosis. The expression of claudin1,3 in the peritoneum may have no change, but the correlation between the peritoneum and the mechanism of EMs need more studies, and it is a new field to study this through claudins.
