Objective:1.To study the relationgship of TAP2 and other correlative immunagenes,HPV16E6/18 E6with Kazakh's esophageal cancer in Xinjiang; 2.To explore the role of the interaction of the TAP2 and other correlative immunity genes,HPV16E6/18E6 and the family history of EC in Kazakh's.Method:1. A 1:2 matched case-control study was conducted with 90 cases of EC and 180 controls. All the cases were collected from the six hospitals in north of Xinjiang during the periods from November 2006 to January 2010. Part of the controls were outpatients without history of malignant tumor, collected during the same period as cases; Another part of the controls were non-esophageal lesions selected from the census on gastrointestinal diseases among the normal population. The controls were matched by sex, nationality, area of residence and age within 5 years difference. To all the studies, the life behavioral factors epidemiological investigations were done and blood samples were collected. We gathered all the cases' paraffin-embedded tissues or fresh tumor tissues and the controls' fresh esophageal mucosa.2. TAP2379 and TAP2665 genetic polymorphisms were identified from the blood samples by PCR-RFLP; and HPV18E6 positive was identified from the paraffin-embedded tissues, fresh tumor tissues and fresh esophageal mucosa with PCR; subjoin the date of HPV18E6,HLA-DRB1*0901.3. All the calculations and statistics were performed with the computer program, SPSS Version 13.0. Statistical analysis adopt 1:2 matchedχ2 test, conditional logistic regression to calculate the values ofχ2, odds ratios and 95% confidence intervals for possible risk/protective factors.Results:1.The frequencies ofTA P2379 genetypein case group is different from the controls group (χ2=11.445, P<0.05,OR:2.714,95%CI:1.521-4.841); The frequencies ofTAP2665 genetype in case group is different from the controls (χ2=4.002, P<0.05, OR:1.735,95%CI:1.011-2.978)。2.The frequencies of HPV16E6 positive in Kazakh's EC is different from the controls group(χ2=11.628, P<0.05, OR:3.133,95%CI:1.625-6.041)。HPV18E6 positive in Kazakh's EC is different from the controls group;HPV18E6doesn'thas same resulrs(χ2=0.018, P>0.05); The frequencies of HLA-DRB1*0901 allele in Kazakh's EC is different from the controls group (χ2=11.505, P<0.05, OR:4.125,95%CI:1.819-9.355)。3.Multivariate conditional Logistic regression analysis showed:Immune-related gene TAP2379 genetic polymorphisms,TAP2665 genetic polymorphisms,HLA-DRB1*0901 allele and biological-factor HPV16E6 could increase the chance of suffering from EC in Kazakh of Xinjiang.The OR were 2.538(95%CI:2.132-4.870),2.197(95%CI:1.205-4.005),3.079(95%CI:1.098-8.639),and 3.302(95%CI:1.535-7.105),4. The interaction analysis show:there were effect-modifications between the TAP2379 genetic polymorphisms and TAP2665 genetic polymorphisms,The TAP2665 genetic polymorphisms and HLA-DRB1*0901 allele,HPV16E6 and TAP2379in Kazakh's esophageal cancer(EC),HLA-DRB1*0901 allele and HPV16E6.Conclusion:1. Immune-related gene TAP2379 and TAP2665genetic polymorphisms,HLA-DRB1*0901 allele and biological-factor HPV16E6 could increase the chance of suffering from EC in Kazakh of Xinjiang.Whether TAP2665genetic polymorphisms were risk factors of EC in Kazakh of Xinjiang needs large case study.2. Family history of EC and TAP2379 genetic polymorphisms,HLA-DRB1*0901 allele and TAP2665 genetic polymorphisms,TAP2379genetic polymorphisms and TAP665 genetic polymorphisms may play a synergy in the course of the development of EC in Kazakh's. HPV16E6 and TAP2379 genetic polymorphisms,HLA-DRB1*0901 allele and TAP2379 genetic polymorphisms,family history of EC and TAP665 genetic polymorphisms,HPV16E6 and TAP2665genetic polymorphisms doesn't play a synergy in the course of the development of EC in Kazakh's.
