Objective: To investigate the polymorphism of methylenetetrahydrofolate reductase (MTHFR) C677T and its association with susceptibility to esophageal cancer (EC) in Kazakh and Han in Xinjiang.Method: MTHFR C677T genotypes were detected by a PCR-based RFLP assay in 179 patients with esophageal carcinoma (Kazakh 94 and Han 84) and 155 population cancer-free controls in the same area (Kazakh 98 and Han 57). Theχ~2 test was adopted to check the difference in genotype distribution between patients and controls, and the accordance with Hardy-Weinberg equilibrium in the group.Result: (1) The frequencies of CC, CT and TT in MTHFR C677T were 56.4%, 36.2% and 7.4% in Kazakh's individuals with EC, respectively. They were significantly different from that in Han's individuals with EC (32.9%,40.0% and 27.1%)(χ~2=15.37,P<0.05);(2) The frequencies of CC , CT and TT were 58.2%,29.6% and 12.2% in Kazakh's controls and 22.8%,52.6%and 24.6% in Han's controls, respectively, a significant difference between them was found(χ~2=18.26,P<0.05).(3) The distribution of C677T genotypes in Kazakh's individuals with EC (CC 56.4%,CT 36.2% and TT 7.4%) was not significantly different from that in Kazakh's controls (CC 58.2%,29.6% and TT 12.2%)(χ~2=1.776,P=0.412).(4) The distribution of C677T genotypes in Han's individuals with EC (CC: 32.9%, CT: 40.0% and TT: 27.1%) was not significantly different from that in Han's controls (CC 22.8%,CT 52.6% and TT 24.6%)(χ~2=2.750,P=0.253).(5) there is no statistical difference between esophageal cancer different grades and MTHFR C677T polymorphism, there is also no statistical difference between transference of lymph node and MTHFR C677T polymorphism.(6) Hardy-Weinberg equilibrium test was performed in the grope and the result showed no statistically significant difference(Hanχ~2=5.086,P>0.05,Kazakhχ~2=1.543,P>0.05)。Conclusion: (1) Distribution of MTHFR C677T polymorphism are different between Kazakh and Han population in Xinjiang. (2) There maybe no relationship between the polymorphism (MTHFR) C677T and risk of esophageal carcinoma in Kazakh and Han in Xinjiang.
