Objective: To investigate the polymorphism of tumor necrosis factor-α-308G/A(TNF-α-308G/A) and its association with susceptibility to Kazakh's esophageal squamous cell carcinoma (ESCC) in Xinjiang.Method: TNF-α-308G/A genotypes were genotyped by PCR-RFLP analysis in 212 paraffin-embedded tissue from Kazakh's ESCC and 200 blood samples from Kazakh's population cancer-free controls in the same area.Theχ~2 test was adopted to check the difference in genotype distribution between patients and controls, and the accordance with Hardy-Weinberg equilibrium in the group.Results: (1)The genotypes of TNF-α-308 G/A are G/G, A/G and A/A.,The frequencies of TNF-α-308 G/A were 70.8%,26.9% and 2.4% in Kazakh's ESCC and were 70%,29% and 1% in Kazakh's controls. There is no significant difference between them(χ~2=1.228, P>0.05).(2)The frequencies of G/G, A/G and A/A in TNF-α-308G/A were 81.7%,18.3% and 0 in well-differentiated EC and were 63.9%,33.0% and 3.1% in moderately differentiated EC and were 58.3%,33.3% and 8.4% in poorly differentiated EC. The genotype distribution of TNF-α-308G/A was significantly different between the different ESCC pathology grading(χ~2=10.630,P=0.019)(.3)The frequencies of G/G, A/G and A/A in TNF-α-308G/A were 61.6%,3.7% and 4.7% in Kazakh's ESCC with lymphaden metastatic and were 77.0%,22.2% and 0.8% in no lymphaden metastatic ESCC. It also revealed statistically significant differences between the TNF-α-308G/A genotype and ESCC lymphaden metastatic(χ~2=7.203,P=0.025).Conclusion: (1) The TNF-α-308G/A polymorphism might not be correlated with the susceptibility to Kazakh's ESCC in Xinjiang.(2)TNF-α-308G/A polymorphism is associated with different ESCC pathology grading and lymphaden metabasis. The G/A and A/A genotype ESCC were tend to poorly differentiated and lymphaden metabasis.
