Iron is one of the important mineral elements in human's body. Iron-deficiency is the ninth of the factors effecting human's healthy. The new trivalent-iron-compound supplement using polysaccharide as ligand, has steady highly water-solubility and can be absorbed as undissociated molecules. Therefore, this compound called polysaccharide iron complex (PIC) is researched more and more.The first part of study includes purring crude tea polysaccharides (27%) and preparation of tea polysaccharides-iron complex (TPS-Fe).After precipitated by ethanol, polyphenol was wiped off by polyvinyl polyrrolidone (PVPP). The optimum reaction conditions were obtained by orthogonal experiment: dosage of PVPP(mTPS:mPVPP) was 5:1, the operation last 0.5 hour and repeated 3 times. The purified tea polysaccharides contained polysaccharide (67%), polyphenol (0.7%), and protein (6.7%). The optimum reaction conditions to prepare TPS-Fe were also obtained by orthogonal experiment: reaction pH was 8, the temperature was 60℃, material ratio was 0.5g:1.2g(mTPS:mFeCl3·6H2O), reaction time was 13h.The crude complex made under the optimum reaction conditions containing 14.6%'s iron, was purified by anhydrous ethanol and aether 3 times respectively and freezed to dry. The production contained 20.17%'s iron. The result of qualitative identification indicated the production did not contains dissociate iron ion. The result of iron dissolution experiment indicated iron in the final production could dissociate quickly and deoxidized by Vitamin C.The second part of this study researched bioavailability in rats of TPS-Fe with FeSO4 as the ferrous sulfate as positive control, according to the method of hemoglobin repletion bioassay. The result indicated: At the end of 21 days'iron supplemented period, values of Hb, total iron in Hb, iron-in-serum of rats from the groups supplemented FeSO4 and TPS-Fe respectively, went up quickly to the normal ones'. There was no significant difference of effect between the dosage of 36 and 48mg/Kg diet supplemented by TPS-Fe. If the bioavailability of FeSO4 was 100% With Hb, total iron in Hb, iron in serum as the metewand, bioavailabilities of TPS-Fe are between 100116%.This data indicated TPS-Fe was prior to FeSO4 as a iron-supplement. According to the research, in some range, values of Hb, total iron in Hb, iron-in-serum, iron contents in blood, heart, livers went up with the addition of dosage of TPS-Fe; content of manganese in blood, value of Cu/Zn in liver went down with the addition of dosage of TPS-Fe; There were no significant relativities between contents of manganese in liver, hearts, values of Cu/Zn in blood, hearts and addition of dosage of TPS-Fe.The third part is a research to pharmacokinetic of TPS-Fe in New Zealand rabbit with Niferex as control. The results insult that, the iron-in-serum-time curve of TPS-Fe fit a two compartment model, the same as Niferex. The maximum iron-in-serum reached to 5.67ug/ml at 4h after priming TPS-Fe. The bioavailabilities in 8h and unlimited time after priming TPS-Fe were 115.84 % and 148.04% respectively.The last part was to research token of TPS-Fe. The results indicated of ultraviolet spectral analysis, Infrared spectral analysis, X-ray diffraction analysis, Fe3+ coordinates with hydroxyl or amidogen. According differential scanning calorimetry and thermo gravimetric analysis to TPS-Fe, it was known that TPS-Fe has good thermo-steadibility.
