ObjectiveFirst trimester spontaneous abortion as one of the common complications of pregnany is a common disease which is caused by many pathology factors and causes early embryonic death. Many factors can cause spontaneous abortion such as mother nuclide endocrine disturbance, autoimmune disorders, uteriue anatomic abnormalities and endometrial infection, and chromosomal abnorality is the most important reason. But there are still many abortions with no definite pathogeny. Recently, it has been shown that the invasive ability of trophoblast cell would become disfunction when spontaneous abortion is caused by many factors. Many studies have proved that the invasive ability of trophoblast cell is mediated by matrix metalloproteinases (MMPs), among which the matrix metalloproteinase-9 (MMP-9) plays major roles. In four tissue inhibitors of metalloproteinases, tissue inhibitor of metalloproteinase-3 (TIMP-3) which plays important roles in the invading process is the important inhibitor of MMP-9. The purpose of this experiment is to detect the expression of MMP-9 and TIMP-3mRNA using RT-PCR in human villi of first trimester spontaneous abortion and discuss the function of MMP-9/TIMP-3 in sustaining the normal early pregnancy.Methods1. Villis were obtained from 30 early spontaneous abortion patients and 20 women requesting termination pregnancy in the First Hospital of ZhengZhou University from July to December in 2007. The average pregnancy were 56-72 days in study group and 48-68 days in control group.2. Total RNA were isolated from villis tissues and the mRNA expression of MMP-9 and TIMP-3 genes were determined using RT-PCR.3. Statistical analysis: data were analyzed by SPSS13.0 statistical software. The quantitative data was expressed by x±s and tested using normal distribution and homogeneity of variance. Independent-samples T test was applied to analyze the difference between groups and 0.05 was defined as size of test.Results1. The expression of MMP-9mRNA in human villi of first trimester spontaneousabortion patients and control groupThrough half-quantitative analysis, the expression of MMP-9mRNA in human villi of first trimester spontaneous abortion is 0.548±0.190, the control group is 0.246±0.140. There is statistical difference between the two groups(P <0.05).2. The expression of TIMP-3mRNA in human villi of first trimester spontaneousabortion patients and control groupThrough half-quantitative analysis, the expression of TIMP-3mRNA in humanvilli of first trimester spontaneous abortion is 0.897±0.101, the control group is0.873±0.114. There is no statistical difference between the two groups(P>0.05).3. The analysis for the proportionality of MMP-9/ TIMP-3mRNA in human villi of first trimester spontaneous abortion patients and control groupThrough half-quantitative analysis, the proportionality of MMP-9/TIMP-3 mRNA in human villi of first trimester spontaneous abortion patients is higher than the control group. The expression of MMP-9/TIMP-3mRNA in human villi of first trimester spontaneous abortion is 0.536±0.266, the control group is 0.191±0.105. There is statistical difference between the two groups(P <0.05). Conclusion1. The expression of MMP-9 in human villus of first trimester spontaneous abortion patients are higher than that in control group, and it hints that the increase of MMP-9mRNA be likely to cause first trimester spontaneous abortion.2. MMP-9/TIMP-3 in human villus of first trimester spontaneous abortion patients are higher than that in control group, which explain the balance of MMP-9 and TIMP-3 play an important role in sustaining the normal early pregnancy.
