Spontaneous abortion is the most common disorder in human pregnancy, according to the statistics, the occurrence of spontaneous abortion after embryo implantation is 31%, 80% are early abortion, Its etiology is complicated,The exact pathogenesis remains unclear by far. In recent years, some evidence has linked peroxisome proliferator-activated receptor-γ(PPARγ) to playing an integral role in the formation and development process of placenta during pregnancy. This review aims to provide a succinct and comprehensive assessment of the role of PPARγ in normal pregnancy and pregnancy complications, and finally its potential as a therapeutic target in the treatment and prevention of adverse pregnancy outcomes.. Obiective:To investigate the expression of PPARγ in villi of the first trimester spontaneous abortion. Methods:To detect the expression of PPARγ in villi from normal human pregnancy(control group,n=40) and spontaneous abortion(experimental group,n=40) during the first trimester of pregnancy by immunohistochemistry, Western Blot, and RT-PCR. Results:1.PPARγ localisation was researched by immunohistochemiscal methods. In control and experimental group, the protein was observed in both villous and extravillous trophoblast nuclei, and in experimental group expression of PPARγ was increased, the difference was statistically significant(P<0.05);2.Western Blot analysis showed that in villi of experimental group, expression of PPARγ was significantly increased than that of control group(P <0.01);3.RT-PCR analysis showed that PPARγ gene was higher expressed in villi of experimental group, and the statistic was significant(P <0.01). Conclusion:1.PPARγ protein was observed in both villous and extravillous trophoblast nuclei. of the control and experimental group.2.PPARγ may play an important role in the modulation of trophoblast invasion, a significantly enhanced protein and mRNA expression of PPARγ was identified in villous and extravillous trophoblast of miscarriage patients may be one factor linked to spontaneous abortion.
