Objective Preliminary study found that the extract of Ocimum basilicum L. can significantly reduce the concentration of plasma TXA2. On this basis, explore the effects of different polar extracts of Uyghur Ocimum basilicum L. on COX-1 and COX-2 activity in vitro. And then, find its anti-thrombus mechanism and its effective parts. Methods①Detected the relative cell vital impulse by MTT colorimetric assay. Screened out the best suitable cell density and administration concentration. And detected the cytotoxicity effect on ECV304 cells and RAW264.7 cells with different extracts. And then determined the IC50 value of each part.②ECV304 cells and LPS-induced RAW264.7 cells were interfered with different polar extracts of Uighur Ocimum basilicum L. in different time. The concentration of 6-keto-PGF1αand PGE2 in supernatant was analyzed by ELISA method. And then calculated the inhibition rate. Results①The best suitable inoculative density of two kinds of cells in 96 well plates is 1×105 /ml. The IC50 values of OBL-petroleum ether, OBL-chloroform, OBL-ethyl acetate, OBL-n-butanol and OBL-ethanol are 62.4, 29.3, 84.8, 258, 320μg·ml-1 respectively; on RAW264.7 cells, followed by the maximum dose concentration of 35, 7.5, 25, 80, 100μg·ml-1; on ECV304 cells, followed by the maximum dose concentration of 50, 7.5, 50, 10, 100μg·ml-1.②The concentration of PGE2 was inhibited by different polar extracts of Uighur Ocimum basilicum L. at different time. The inhibition ratio by acetidin part and n-butanol part were higher than other parts, which maximal inhibition ratio in short term were 53.95% and 46.02% while in long term were 53.93% and 53.41%. Different extract parts had different effects on the concentration of 6-keto-PGF1αat different time. OBL-n-butanol and OBL-ethanol significantly increased the concentration of 6-keto-PGF1αin the short term which inhibition ratio in 1 h were -56.85% and -98.17%, while in 4 h were -48.72% and -50.20%. In the long term, the maximal inhibition ratio of OBL-ethyl acetate was 19.68%. Inversely , OBL-chloroform and OBL-n-butanol increased the concentration of 6-keto-PGF1αsignificantly which minimal inhibition ratio were -33.79% and 38.71%. Conclusion Uighur Ocimum basilicum L. may inhibit the enzymic activities of TXA2 synthetic enzyme and activate the enzymic activity of COX-1. Ultimately, the balance between TXA2 and PGI2 is maintained which can cause anti-thrombus. It may also inhibit the enzymic activities of COX-2 which lead to the degrading of PGE2, and then cause anti-thrombus. And the ethyl acetate and n-butanol extract parts are the effective parts when to play the role.
