Objective: To study the preventic and therapeutic action that extract formula of plant-metallothionein on cadmium poisoning rats, and to provide a theoretical basis for exploring a new type of heavy metal poisoning antagonist– metallothionein. Methods:60 Wistar rats were used and allocated into six groups: negative control group; positive control group; preventic-one group(low-dose MT preventic group); preventic-two group(high-dose MT preventic group); therapeutic-one group(low-dose MT therapeutic group); therapeutic-two group(high-dose MT therapeutic group); CdCl2 was used to establish cadmium poisoning model, and extract formula of plant-metallothionein was used to interfere: preventic groups were used two weeks before CdCl2 was used, therapeutic groups were used two weeks after CdCl2 was used. The content of MDA and activity of SOD, GSH-Px and CAT in liver , serum and kidney and content of IL-1β,IL-6 in serum was detected to analyze the preventic and therapeutic action of the formula on cadmium poisoning. The Ultrastructural changes of rats in kidney and liver were observed. Results:1) GSH-Px, SOD activity and the content of MDA in liver of preventic and therapeutic groups had significant difference with cadmium poisoning group (P<0.05); 2) GSH-Px, SOD activity and the content of MDA in kidney of preventic and therapeutic groups had significant difference with cadmium poisoning group (P<0.05); 3) GSH-Px, SOD activity and the content of MDA in serum of preventic and therapeutic groups had significant difference with cadmium poisoning group (P<0.05); and MT content of high-dose MT preventic group had significant difference with therapeutic groups (P<0.05); 4) IL-1βand IL-6 content in serum of preventic and therapeutic groups had significant difference with cadmium poisoning group (P<0.05); Conclusion:The excessive amount of cadmium chloride can promote the enhance of animal lipid peroxidation and inhibit the activity of GSH-Px, SOD and CAT, and can inhibit the immune action, decreace the IL-1βand IL-6 level. Animal experiments showed that extract formula of Xinjiang plant-metallothionein had a certain antagonism against cadmium poisoning and could reduce lipid peroxidation and immune injury and the inhibited action to GSH-Px, SOD activity, and the earlier used ,the effect was more significant...
