| Objective:Observing the human stomach cancer cell of inhibitory effect of WEI-AI-PING and alteration of activity of enzymes of cancer cell,and the apoptosis induced by WEI-AI-PING to study the effects and mechanism of WEI-AI-PING on the apoptosis of Human Stomach Cancer SGC-7901 cells and to provide reliable evidence to future application to clinical practice.Methods:In Vitro,the Human Stomach Cancer cell Line SGC-7901 were cultured.The growth inhibitory rate was evaluated by MTT assay.Morphological changes of apoptosis of human stomach cancer SGC-7901 cells was observed by light microscopy and apoptosis of human stomach cancer SGC-7901 cells was investigated by electron microscopy.the alteration of activity of enzymes of cancer cells was observed by applying the enzyme cytochemical method.Results:(1) MTT method reporte: compared with control group,the three groups of serum contration of WEI-AI-PING,the group of DDP and Chinese and western medicine group all are proved to be statistically significant on growth inhibitory rate after 24 hours'culture.(2)There are obvious changes of morphology of the SGC-7901 cells as the loss of normal morphology,spindle cells becoming round,cytoplasmic concentration,nuclear shrinkage in the light microscopy (3)the observation of the electron microscopy shows the formation of the apoptotic cell and apoptotic bodies can be observated by the electron microscopy.(4)Enzyme cytochemical method indicates that the response granules of G-6-Pase and Mg2+-ATPase in the colon cell become smaller after the treatment of serum contained Chinese herbs, the amount the granules become fewer,the density is lower, some are negative. The activity of the enzyme decreased obviously.Conclusion:(1)WEI-AI-PING has the function of inhibiting the proliferation of the human stomach cancer cell line SGC-7901. (2) )WEI-AI-PING can decrease the activity of the G-6-Pase and Mg2+-ATPase of the human stomach cancer cell line SGC-7901.(3) WEI-AI-PING can induce the cancer cell apoptosis (4) WEI-AI-PING can inhibit human stomach cancer cells'ability of invasion and metastasis.(5)the experimental research reveals the TCM-Western Medicine integration shows better inhibit effect of cancer cell and it will be an good attempt to treat cancer in clinicai practice.(5) inhibit prolife ration of the human stomach cancer SGC-7901 cells,decreasing the activity of the G-6-Pase and Mg2+-ATPase, inhibiting invasion and metastasis and inducing apoptosis may be the main mechanism of Anti-tumor of WEI-AI-PING. |
