Purpose: To study the effects of YI-CHANG-TAI on proliferation and activity of enzymes of colon cancer cell and explain the mechanism of its resisting-cancer function.Methods: In vitro, we cultured the cell strain of the human colon cancer cell CCL229. The inhibition of cell proliferation was determined by MTT method, the activity of G-6-Pase and Mg2+-ATPase was detected by enzyme cytochemistry, and the apoptosis was determined by light microscopy, electron microscopy.Results: (1) The serum contained Chinese herbs has evident inhibitory function to the human colon cancer cell CCL229, and the inhibitory effects have time dependence. The inhibiting rate is 54.78% after 24h culture, 65.6% after 48h culture. (2) The observation of LM shows the morphological change of the colon cells of Chinese herbs group. (3) The observation of EM shows the formation of the apoptotic body, which is the most characteristic change of the apoptosis cell. (4) The observation of EM also shows the decreased amount of the microvilli, which play a key role in the process of invasion and metastasis. (5) Enzyme cytochemistry method indicates that the response granules of G-6-Pase and Mg2+-ATPase in the colon cell become smaller after the treatment of serum contained Chinese herbs, the amount of the granules become fewer, the density is lower, some are negative. The activity of the enzyme decreased obviously.Conclusions: (1) YI-CHANG-TAI has the function of inhibiting theproliferation of the human colon cancer cell CCL229. (2)YI-CHANG-TAI can induce the human colon cancer cell apoptosis. (3)YI-CHANG-TAI can decrease the activity of the G-6-Pase andMg2+-ATPase of the human colon cancer cell. (4) YI-CHANG-TAI caninhibit human colon cancer cells' ability of invasion and metastasis.
