| Object At present, the practice of lung transplantation is constrained by a shortage of suitable donor organs. So new strategies to prolong effectively the reliable duration of lung preservation .The studies used Kyoto-1 solution for different times and at different temperature to observe the activities of succinate-dehydrogenase (SDH) and alkaline- phosphomonoesterase (ALP) and provided basic theory for preserving donor and practically useful indicator to detect the well reserved lung. Methods Using Kyoto- 1 solution, lungs preserved for 4h, 1 2h, 24h and 48h at 4 0C or 0 0C, respectively. With cytochemistry technique, the activities of ALP and SDH were detected; Under light microscope, the distribution of activities of ALP 5 and SDH between normal control group and experiment groups were observed; Using Microimage analysis system measured the activity of enzyme and using the method of average and standard deviation treated the data; Under EM, the distriction, quantitatively analysis and the ultrastructure changes. Results 4 Under light microscope, ALP distributed mainly in alveolar type II cell and terminal bronchiolar mucous epithelia cells. To lungs preserved for 4h, 1 2h at 4 0C, 24h at 4 or 4h, 12h at 00C, the activity of ALP didn抰 change obviously (p>O.05) compared with normal control groop .To lungs preserved for 24h at 00C ,the activity of ALP decreased obviously .To lungs preserved for 48h at 0 0C and 4 0C ,the activity of ALP decreased significantly (p0.05).To lungs preserved 6 for 24h at 0 0C ,the activity of SDH decreased obviously (p |
