Objective: At present, there exist many kinds of liver preservation solutions,most ofwhich have their own defects. Through using PV liver preservation solution and making acomparison with UW liquid preservation solution which is regarded as "gold standard", itaims to research into the effect of the solution on preservation of rat liver and the survival effectafter rat liver transplantation, which makes a good foundation for further clinicaltrials.Methods:Number of240SD rats will be randomly divided into PV fluid groupand UW solution group. Depending on the different monitoring indicators, they willbe divided into the following groups: PV fluid group and UW solution group aredivided into6h,12h,18h and six subgroups1h,12h after liver transplantation.Each subgroups includes6pairs of rats, adopting the traditional two cuff method toestablish the model of liver transplantation. The experiment will observe and test liverenzyme learn (ALT, AST and LDH), the concentration of malondialdehyde (MDA),superoxide dismutase (SOD), cell apoptosis, bile secretion, organization morphology.In the second group, PV fluid group and UW solution group are divided into threesubgroups6h,12h, and18h. Each group includes eight rats. It will focus on theobservation on survival rate7days later after rat liver transplantation.Results:From the comparison between PV fluid group and UW solution, it wasfound that rats blood enzymology had little significant difference (P>0.05), theorganization structure change under light and electron microscopy was similar. Afterthe preservation of12h, MDA concentration in PV group is significantly lower than UWsolution group (P <0.05), while18h, function of bile secretion in PV group is slightlyweak than in UW solution group (P <0.05). There is little significant difference insurvival rate7days after rat liver transplantation (P <0.05).Conclusion:PV fluid and UW solution both have similar preservation effect inrat liver preservation. PV liquid is slightly better than the UW solution in scavengingoxygen free radicals and antioxidant.
