Object i ye To examine the inhibition effect of Epidernial growth factor receptor(EGFR) antisense RNA on hepatocarcinoma cell line SMMC-772 1 and explore the significance of antisense RNA therapy for of liver cancer.Mater i a Is and Methods Eukaryotic expression vector of EGFR antisense RNA, pLXSN-AE5? was transfected into SMMC-7721 by using Stearylamine/DOPE (SA liposome). The cells were selected with G4 18 and the total number of formed colonies was counted. The growth rate and the ability of the transfected cells to form colonies in soft agar medium were also examined. Meanwhile, cell cycle was examined by flow cytometry.Resu I ts OAntisense RNA was expressed in the transfected cells by RT-PCR methods. 甌he growth of antisense transfected cells was prominently suppressed comparing with SMMC-772 1mimic transfected with SA liposome and empty vector transfected cells, respectly. The double time of antisense transfected cell was significantly delay (beyond 8 days) than SMMC-772 1. SA liposome and vector transfected cells (4-5 days). 甌he number of colonies formed in soft agar by SMMC-7721 and transfected cells (SA liposome~ pLXSN and pLXSN-AE5? was 254.8?15.9, 23 1.3?18.6, 246.3?3.2, 54.2?.3 respectly. The antisense transfected cells partly lost the ability of anchorageindependent growth in soft agar. ~The percentages of cells in S phase of SMMC-7721 treated with SA liposome.. pLXSN vector.. pLXSN-AE5?or SMMC-7721 was 68.34%.. 71.3%.. 26.81% or 67.43% respectly. The percentages in G0-G1 was 18.58%-. 16.43%59.10% or 19.41%.Conc I us i on 〦GFR antisense gene effectively reverse the malignant phenotype of human hepatoma cells; ?EGFR antisense gene is of potentiai clinical utility in treatment of hepatocellular carcinoma.
