The effective fractions of Forsythia suspense(Thunb.)Vahl are founded to be able to anti influenza Virus.The dissertation divides into four parts to study the material of anti-influenza Virus of Forsythia suspense(Thunb.)Vahl.Part one: Literature and summarizations about Forsythia suspense(Thunb.)Vahl.It sums respectively up the pharmacy, pharmacology of Forsythia suspense(Thunb.)Vahl, 67 references are cited.Part two: Experiment studies about Forsythia suspense(Thunb.)Vahl.One: The observation of the extracted term of Forsythia suspense(Thunb.)Vahl with the evaluation quota of the amount of the total phenols and phillyrin give the result that the best extracted term is:The drug is extracted by 50% alcohol for three times, each time 8 times 50% alcohol and one hour.Observing the absorption faction with the evaluation quota of the amount of the total phenols and phillyrin of Forsythia suspense(Thunb.)Vahl on the five kinds of macro absorption resins.We reach the conclusion that the AB-8 resin has the best macro absorption resins which has the best effect on the separation of the total phenols and phillyrin of Forsythia suspense(Thunb.)Vahl and the best technology term,The best technology term is that the 20mg/mL density of the extraction substance is passes the macro porous resin column of d/L is one to eight with the 1mL/min absorbed speed,and water is used as elution until the result of the reaction -Naphthol/thick sulphuri acid is negative.Then elute it with 50% alcohol for ten times of resin column with the 1mL/min eluted speed.This method is simple and we can put it into practice.Two: The effects of anti-influenza Virus from Forsythia suspense(Thunb.)Vahl are extracted and separated.15 compounds are isolated with Silica gel, Polyamide, Sepadex gel,Toyopearl gel chromatography and 10 compounds are identified with UV,IR,EI-MS,1HNNR,13CNMR,FAB-MS spectra.They are four caffeol glycosides(Forsythoside A,Forsythoside D,Forsythoside C,β-Hydroxyacteoside), one flavonoids (rutin),two ligan glucosides((+)-phillyrin,(+)-pinoresinal-β-D-glucoside),two organic acid(Caffenic acid,Ursolic acid),and one sterol(β-sitosterol). All of the 10 compounds are firstly obtained from the effective fractions of Forsythia suspense(Thunb.)Vahl,Especially,β-Hydroxyacteoside is obtained from Forsythia suspense(Thunb.)Vahl for the first time.Three: The total phenols contents in the effects of anti-influenza Virus from Forsythia suspense(Thunb.)Vahl are determined with the evaluation quota of Forsythoside A and rutin respectively.They are 28.53%,91.07% respectively.The calibration curve are linear at ranges of 0.022-0.66ug(for Forsythoside A, r=0.9998 ),9.088-81.29ug/mL(for rutin, r=0.9998) .The average recoveries are 97.97%(RSD 1.42%,n=5) and 98.35%( RSD 1.28%,n=5).The contents of index compounds including Forsythoside A,Phillyrin and rutin are determined by HPLC.The calibration curve are linear at ranges of 0.1515-1.01 ug(for Forsythoside A,r=0.9998),0.24-1.2ug(for phillyrin,r=0.9998),and 0.15-0.60ug(for rutin,r=0.9997) respectively.The average recoveries are 100.79%(RSD 2.91%,n=5),99.00%(RSD 2.53%,n=5) and 99.46%( RSD 2.99%,n=5) respectively.Part three: The determination of crocetin in effective fractions Ⅰand Ⅱ from Huanglianjiedu Decoction were established by HPLC and TLC.Using the HPLC,the linear range of crocetin is 0.0751-0.3140ug(r=0.9995),the contents of effective fraction Ⅰand Ⅱ are 0.0767% (RSD2.08%,n=3) and 0.3383%(RSD1.67%,n=3) respectively,The average recovery is 101.68%(RSD 1.16%,n=5) .Using the TLC, the linear range of crocetin is 0.0628-0.1884ug(r=0.9996),the contents of effective fraction Ⅰand Ⅱ are 0.0779% (RSD 2.82%,n=3) and 0.3510%(RSD1.40%,n=3) respectively.The average recovery is 98.54%(RSD 0.83%,n=5) .The two methods are simple and accurate and could be used to control the quality of the effective fractions from Huanglianjiedu Decoction.Part four: analysis and discussion of experiments...
