Objective:To study the expression of the vascular endothelial growth factor –A,-C,-R3mRNA and Protein and the relation among the expression of the three factors with cervical lymph nodes metastasis in laryngeal squamous cell carcinoma(LSCC)and to investigate the mechanism of development and lymphatic metastasis.Methods:VEGF-A,-C,-R3 gene protein in 50 cases of laryngeal carcin- oma and 15 cases of normal tissue around the cancer were detected by immuno- histochemistry. VEGF-A,-C,-R3mRNA in 30 cases of laryngeal carcinoma and 15 cases of normal tissue around the cancer were examined by reverse transcription polymerase chain reaction(RT-PCR). To study the relation between the three factors with clinicopathological parameters and lymphatic metastasis by two experiments,the laryngeal carcinoma were devided by age,T tage, phathologic stage and lymphatic metastasis . Results: Immunohistochemistry and RT-PCR both show:Expression of VEGF-A,-C,-R3 was significantly higher in laryngeal carcinoma than in normal tissue around the cancer. Expression of VEGF-A were correlated with T stage and the pathologic stage,nor with age ,but expression of VEGF-C,-R3 were not associated with T stage, age and pathologic stage. The overexpression of VEGF-R3 had a positive correlation with the higher expression of VEGF-C. The overexpression of VEGF-A,-C,-R3 was related to cervical lymph nodes metastasis in two experiments,and significantly statistical differences exist between the quantitative expression of three factors in the lymph nodes metastatic group and in each no lymph node metastatic group of different clinic type.Conclusion:1.The overexpression of VEGF-A,-C,-R3 in laryngeal carcinoma was related to cervical lymph nodes metastasis ,which suggested that they would become a biologic indexes to prognosis of laryngeal carcinoma.2. VEGF-A,-C,-R3 were overexpression in laryngeal carcinoma,which suggested that they would be one of the reasons of development and progression of LSCC.3.The overexpression of VEGF-C was related to VEGF-R3, which suggested that they would be up each other.
