| Objective:To probe channel and point- entrails correlation was one of the most important content of revealing meridian essence and researching acupuncture mechanism of action. Considerable clinic and empirical study indicated that Neiguan cave had distint effect to treat is chemic heart diease. The method of this experiment was to investigate The rabbit's ST potential of ECG of acute myocardial ischemia (AMI) rabbits,and to investigate the pathophysiological change of the content of endothelins (ET), thromboxane B2 (TXB2) and 6-keto- prostaglandinfla (6-Keto-PGF1a),the latter both of which is the stably metabolic products of TXA2 and PGI2 ,in plasma of AMI rabbits. To study the therapeutic effective of electroacupuncture(EA) on AMI model of rabbits and from the scope of adjusting ET,TXA2 and PGI2 ,the vasoactive substance origined from endothelio ,to study the possible functional pathways. To explain furtherly correlative mechanism of action about Neiguan-Heart. All this provide the basic theory for the clinic.Methods:Experiments were performed on 32 big ear rabbits of Japan,whose body weight was from 1.8kg to 2.3kg and whose female or male was not limited. they were randomly devided into four groups:normal group,notho-operation group,model group and EA group. Normal group were not provided treatment. Model group were made by ligating ramus descendens anterior arteriae coronariae sinistrae to make the AMI model. Notho-operation group only opened breast to expose heart and did not ligated but to cut through a thread. EA group was made by Ren's method. The rabbit's ST potential of ECG was recorded in every needed time. At 30min and 60min after the model was made, ET,TXB2 and 6-Keto-PGF1a level in plasma were dynamically measured by radioimmunoassay and analyzed their dependablity.Results:The rabbit's ST potential of ECG of the model group and EA group significantly step up than that of the normal group and notho-operation group at 30min after the model was made. But at 60min after the model was made, The rabbit's ST potential of ECG of the EA group can evidently reduced,but that of the model group was still super . Every index of notho-operation group was not significant deviation from that of normal group (P>0.05). ET level of model group was significantly higher compared to notho-operation group and normal group,so did TXB2 and TXB2/6-Keto-PGF1a (P<0.05 or P<0.01) . However 6-Keto-PGF1a level was significantly lower compared to notho-operation group and normal group (P<0.01). In EA group ET level was cut down,so did TXB2 and TXB2 /6-Keto-PGF1a. However 6-Keto-PGF1a was stepped up. Moreover,ET level was favourable positive correlation to TXB2 /6-Keto-PGF1a. The experiment implied that EA could obviously improved AMI.Conclusion:EA on Neiguan can evidently reduce the preternatural increase of ST potential of ECG. The rising of ET and TXB2 levels and the decreasing of 6-Keto-PGF1a level of plasma in AMI model confirmed that vasoactive substance origined from endothelio were positively correlated with the degree of AMI. The favourable positive correlation about ET level and TXB2/6-Keto-PGF1a confirmed that ET,TXA2 and PGI2, those who could aggravated the damage of AMI in common. These datas suggested that ET,TXA2 and PGI2 may play an important role in AMI. EA effective by two side Neiguan could regulate the plasma level of ET, TXA2 and PGI2 to improve AMI and potect the heart. This may be impossible functional pathway of EA on AMI model. This experiment also confirmed that the basic theory of favourable correlation of Neiguan-heart from the scope of the humoral factor. |
