Objective:To study the effects of electroacupuncture at “Neiguan”(PC 6)on specific metabolites and metabolic pathways in the myocardial tissue in rats with myocardial ischemia reperfusion injury(MIRI)based on 1H nuclear magnetic resonance(1H NMR)by screening specific metabolites,and compare the specificity electroacupuncture at “Hegu”(LI4)in the prevention of myocardial ischemia reperfusion injury,so as to reveal its mechanism underlying protection of electroacupuncture on MIRI from the aspect of metabonomics.Methods: 48 male SD rats were randomly divided into blank control,model,EA“Neiguan”(PC6)and EA “Hegu”(LI4)groups(n=12/ group).The Control group was banded on rat board for 30 min,once daily foy 7 days.The model group was banded on rat board as the control group until the 7th day and ligated the left anterior descending coronary artery for 40 min,restored the blood flow for 1 h to form the MIRI model.EA groups(10Hz/50 Hz,1m A):To electroacupuncture bilateral “Neiguan”(PC 6)and “Hegu”(LI4)for 30 min,once daily foy 7 days,to 7th day form the MIRI model.Collecting the myocardial tissue of rats in each group.myocardial tissues assay the profiles of differential metabolites using 1H nuclear magnetic resonance(1H NMR)spectroscopy and multivariate statistical analysis as the principal component analysis(PCA),projection to latent structures discriminate analysis(PLS-DA)and orthogonal projection to latent structures discriminate analysis(OPLS-DA),so as to seek specific metabolites by 1H nuclear magnetic resonance(1H NMR).Results :1.Compared with the control group,the contents of myocardial tissue fumarate (P<0.001)in model group were significantly increased,while leucine(P<0.05),propanediol(P<0.001),3-hydroxy butyricacid(P<0.001),glutamate(P<0.001),glycerol phosphocholine(P<0.001),aspartate(P<0.01),inosine(P<0.01),lactate(P<0.001)were significantly decreased.These changes in content were statistically significant,which are the potential biomarkers of myocardium.2.Compared with the model group,the contents of myocardial tissue propanediol(P<0.05),leucine(P<0.05),acetate(P<0.01),phosphocholine(P<0.05),ni acinamide(P<0.001),Glycerol phosphocholine(P<0.01),inosine(P<0.05),glucose(P<0.001)in EA “Neiguan” group were significantly increased,while succinate(P<0.01),adenosine nophosphate(AMP,P<0.05),ncotinamide adenine dinucleotide(NAD+,P<0.05)were significantly decreased.3.EA “Neiguan” was able to map the metabolite profile of myocardial tissue of MIRI rat to control group.4.Compared with the model group,the myocardial tissue metabolic patterns of“Hegu”group were indistinguishable,indicating that EA “ Hegu” did not change the myocardial tissue metabolic patterns in rats.Conclusion:The myocardial tissue of MIRI has obvious metabolic characteristics,and metabolic pathways such as glucose metabolism,energy metabolism,amino acid metabolism,ketone metabolism were affected by changes in metabolite content,indicating that electroacupuncture at “Neiguan” have a benign adjustment and preprotective effect.Electroacupuncture at “Hegu” without preprotective effect on the myocardial tissue,which indicates that there is a special relationship between “Neiguan” of the pericardium channel and the meridians viscerat,as well as the specific of acupoint.
