Centipeda minima (L.) A.Br.et Ascher. is an annual herbaceous plant of Compositae,which has been used as a fine remedy for cold,headache.chronic nasal allergy and cough in southeast Asia.Recent pharmacological study has proved it has inhibitory activity agaist bacteria,allergy and tumor etc.In our phytochemical study of this plant,we screened out a active constituent,which showed significant antitumor activity.The extraction and isolation from the whole plants of Centipeda minima were accomplished and 31 constituents were identified. 8 components were isolated from Et2O fraction and EtOAc fraction,5 of them were identified as follows:brevilin( I ), quercetin-3-methyl-ether( Ⅱ ), quercetin-3,3' -dimethyl-ether(Ⅲ), 3,3 ' ,5,5' -tetramethoxy-trans-stilbene( Ⅳ ), 2-isopropyl-5-methyl-p-hydroquinone-4-O- β -D-xylopyranoside(Ⅴ).Essential oils were extracted by steam distillation(SD) and analyzed by gas chromatography-mass spectrometry.26 constituents were identified as follows:trans-chrysanthenyl acetate(Ⅸ) , thymol(Ⅹ) , maaliene(Ⅺ) , isocomene(Ⅻ), [2,6,6-Trimethylcyclohex-1 -enylmethanesulfonyl]-benzene(ⅩⅢ), [7,2,0]undec-4-ene-4,11,11-trimethyl-8-methylene-[1R-(1R*,4Z,9S*)]-bicyclo(ⅩⅣ ),2-methyl-3-methylene-2-(4-methyl-3pentenyl)-(1S-endo)-Bicyclo-[2,2,l]-heptane (ⅩⅤ), a-Caryophylene(ⅩⅥ), 5-methyl-2-(1-methylethyl)-acetate-phenol(ⅩⅦ), 2-methyl-3,7-dimethyl-2,6-octadienylester-propanoic(ⅩⅧ),isocaryophillene(ⅩⅨ) etc.23 compounds were discovered for the first time from Centipeda minima (L.) A.Br.et Ascher.Under the guidance of pharmacological experiment and analysis by FCM,we researched the active fractions and constituents.As a result,ether fraction was most active. A constituent was isolated from this fraction and was identified as brevilin.It clearly showed significant inhibitory activity agaist pancreas,BEL-7402 and MDA-MB-231 tumor cell,the IC50 were 11.6μg/ml,2.0μg/ml and 3.6μg/ml.Compared the extracts of SFE and SD,it clearly showed that the forrner was more active.Applied GC-MS,two of the extracts were contrasted and analysed.As aresult,there were 11 same components.In the extract of SFE,50 peaks were isolated and 21 of them were identified,account for 64.37%.The content of glabelin is most abundant,this kind of sesquiterpene lactone has antitumor activity.In the extracts of SD,50 peaks were isolated and 26 of them were identified,occupied 84.65%.The content of trans-chrysanthenyl acetate is most abundant,this kind of constituent has no active.the result proved pharmacological experiment.On the base of antitumor constituent of Centipeda minima,we established its quality standard systematically.The TLC was unique to some extent and the quantitative analysis was highly sensitive,well repeated and promisingly reliable.Our research had enriched quality standard of Centipeda minima in Chinese Pharmacopoeia(2000) and could control its quality effectively.
