Chronic skin ulcer (CSU) is a common clinical disease which can be secondary to many diseases and be treated difficultly. Collagen metabolism being out of control is an important factor in CSU. Excessive collagen degradation and depressed collagen synthesis ability lead to unhealing wound. There are four kinds of traditional Chinese medicine (TCM) often being used: removing carrion medicine, promoting blood flow medicine, warming yang medicine and stypsis medicine. Clinical practice and animal experiment both proved that topical used TCM had preferable curative effect on CSU. We hypothesized that these four kinds of TCM may have effects on the collagen metabolism of human skin fibroblasts (HSFb). In this experiment, we cultured HSFb as research object and choosed main components of several representative topical used TCM as tested drugs to study the mechanism of the effects of these four kinds of TCM on CSU. These components are soluble pearl powder (SPD, stypsis medicine), dracohodin perochlorate (DP, promoting blood flow medicine), boswellin aqueous extract (BAE, promoting blood flow medicine), velver antler polypeptide (VAP, warming yang medicine) and arsenic trioxide (AT, removing carrion medicine).This paper has two parts. The first one is composed of two reviews including collagen metabolism and CSU, and current situation of therapeutics of topical medicine for CSU. Then on the basis of these studies, we carried out this experiment.1.The effects of tested drugs on proliferation and collagen synthesis of HSFb.Method: MTT assay was used for observing cell proliferation and hydroxyproline for collagen synthesis.The results showed that:(1)After cultured with the tested drugs for 48 hours, the proliferation rate of HSFb were elevated by SPD, DP, BAE, VAP and lowered by AT. For SPD, DP, BAE and VAP, obvious proliferation-promoting concentrations on HSFb were 1.25×102~1×103μg/ml, 5~20μg/ml, 0.64~2×103μg/ml and 6.25×10-3~0.4μg/ml, respectively (p<0.01). In concentrations of 0.16~0.8μg/ml, AT lowered the proliferation rate of HSFb.(2) After cultured with the tested drugs for 48 hours, collagen synthesis of HSFb were elevated by SPD, DP, BAE, VAP and lowered by AT.
