To explore the crude polysaccharide bioactivity in Curcuma zedoaria Rosc., we study the best isolating Conditions and optimize the isolating techniques. Crude polysaccharide was gained by hot water extraction, alcohol precipitation, decolorization and deproteinization. Then, with 50% and 75% alcohol respectively, divided it into two fractions: CZ1 and CZ2. We did chromatography with DEAE-cellulose column for each fraction, and compared the differences of chromatographic eluate curves by adjusting pH of DEAE-cellulose column. CZ1 was a relatively pure polysaccharide, and CZ2 was divided into two fractions when in pH5.5 and pH7.0, and only one fraction when in pHlO.O. With the increase of pH, reclamation rate goes down and polysaccharide content goes up. It is showed that pH was an important factor, and the best pH condition for isolating polysaccharides in Curcuma zedoaria Rosc. was 7.0.The bioactivities of immunology, anti-oxidative function and antitumor activity of the polysaccharides in vivo or vitro were studied. The results showed that a higher antitumor S180 effect of the polysaccharides by injection was observed. The most inhibitive rate was 63.19% when the dose at 500mg./kg·day. The Curcuma zedoaria Rosc. polyszccharide had a evident effect in eliminating ·OH and O2-, and their the biggest eliminating ratio was 88.91% and 98.5% respectively; SOD activity and LPO content were significantly changed, SOD activity was enhanced 145% and LPO content reduced 40.95%, maximally, comparing with the control group; The weights of all chickens were increased quickly from the second week after treatment, the biggest increase weight was 48.75g per week; The phagocytosis ratio and index of blood corpuscle, weight and growth speed of bursa F and spleen in group of Curcuma zedoaria Rosc. polyszccharide were increased; The values of antioxidant and immunity functions in the group fed with the cyclophoshamide added Curcuma zedoaria Rosc. polyszccharide were similar to the control group, which it showed that the injury to immune organs by cyclophoshamide could be made up by Curcuma zedoaria Rosc. polyszccharide. All in all, Curcuma zedoaria Rosc. polyszccharide could eliminate ·OH and O2-, increase SOD activity, and reduce LPOcontent. Furthermore, it had a antioxidant ability in some degree, could increase growth performance of chickens, repair injury of tissue, strengthen body immunity.
