Objective: To provide molecular evidences for its breeding by studying the genetic relationship among varieties of Bupleurum baokang (Bupleurum chinense DC.)Methods: A total of thirteen samples including twelve Bupleurum baokang isolated from four areas and one Bupleurum falcatum L. were obtained from Maqiao town of Hubei Province. The radix of each sample of Bupleurum baokang were identified by characteristics and microscopical characters and detected by chemical characters. DNA was extracted from leaves of all the samples and analyed by Randomly Amplified Polymorphic DNA (RAPD) method. To calculate Jaccard measure and draw dendrograms by using genetic distance UPGMA method.Result:(1)On the characteristics of radix, Bupleurum baokang is similar to Bupleurum chinense DC, and minority of them are different on the outline and the amount of branches to Bupleurum chinense DC.(2) Bupleurum baokang is identical with Bupleurum chinense DC. on the microstructure.(3) Bupleurum baokang and Bupleurum chinense DC. are identical on chemical characters.(4)The RAPD analysis showed that there were a significant genetic diversity among the 13 samples. A total of 61 bands was amplified by the 7 primers, 95.1% of the total bands was 58 polymorphic, 4.9% of the total bands was 3 monomorphic. Of the four populations within Bupleurum baokang, the level of RAPD polymorohism is from 33.3% to 78.1%.The genetic similarity(GS) between Bupleurum baokang and Bupleurum falcatum L. was from 0.209 to 0.333. The GS of each population was from 0.267 to 0.857. The degree of similarity of three samples of GAP population was the highest in four populations of Bupleurum baokang.The results of cluster analysis by using UPGMA method showed that all the tested accessions can be differentiated by RAPD markers, and classified as 3 groups. Of which, two groups of Bupleurum baokang were classified as one group.Conclusion: Bupleurum baokang was is similar to Bupleurum chinense DC. on characteristics, microstructure, chemical characters and the contents of saponin and actually existed much genetic diversity on molecular level among the germplasm resources.
