| HongJingtian is the dry root and rhizome of Rhodiola crenulata (Hook.f.et Thoms.)H.Ohba.belongs to Crassulaceae.HongJingtian has a complicated origin in our state market and it's quality varies greatly. To ensure the using of of clinical drugs and medicine is safe and effective,we must identificate HongJingtian herb; To clarify Rhodiola pharmacodynamics material,we must study its chemical components; The quality analysis of HongJingtian is restricted for the reason of Salidroside is a single reference index, Inevitably we must determine Multiple Indicator; Traditional identification of HongJingtian has limitations, so it is difficult to identificate similar varieties, drug powder and proprietary Chinese medicine, It is significantly to establish a strong, reliable, simple, and rapid modern methods of identification.The dissertation includes two parts: Summary of related literatures and Experiments.Part I: Summarized the related literatures of Rhodiola root in habitat, sources and identification, chemical components, pharmacological action, and quality analysis; Summary of infrared spectroscopy in Quality Evaluation of Chinese medicine identification. 128 reference were be cited.Part II: Experiments. It has four chapters. The first chapter, the identification of HongJingtian. The second Chapter, Studies on chemical constituents of Rhodiola crenulata; The third Chapter ,The quantitative determination of active components. The four Chapter , IR identification in Rhodiola. The results as follows :The first Chapter, Indentificated the Hongjingtian and classified drug into two part : stem feet and roots. All these lay the foundation of accuracy and reliability of study on the chemical composition of Rhodiola crenulata and quality evaluation.The second Chapter, Studies on chemical constituents of Rhodiola crenulata. Several purification and separation methods and many kinds of structure elucidation and identification technologies, such as UV, IR, EI-MS, ESI-MS, 1HNMR, 13CNMR, HMQC were used. 7 components were separated from Rhodiola crenulata root and rhizoma,and 6 compounds were identified which included Caffeic acid, Vanillin, Salidroside, Gallic acid, Daucosterol, Tricin-7-O-β-D-glucoside.Vanillin was isolated from Rhodiola.s for the first time. Tricin-7-O-β-D-glucoside was isolated from Rhodiola crenulata. for the first time.In addition, compounds ZMP-2 is structural of nuclear-cinnamoyl. Its sugar part is the seven carbon sugar .According to data from the literature spectroscopy inspection, no similar structure has been found before.It may be a new compounds.The third Chapter ,The quantitative determination of active components. Sample extraction : Rhodiola 0.5g, 20ml methanol, 45 min and chromatography for detection: identify ways to extract powder medicine ultrasound to determine Chromatography conditions for the detection wavelength of 223nm. The mobile phase was 0.5% phosphoric acid water-methanol (80:20), a flow rate of 1 ml min-1. The injection volume was 5μl. The determination of three components salidroside and tyrosol, gallic acid is established by HPLC methods. The method is simple, accurate, reproducible good recovery qualified, accurate results.,it can be used as the quality control methods for the development of scientific quality evaluation.Analysis showed that Rhodiola crenulata different parts of the effective components in the stem shoot higher than rhizomes; Rhodiola storage life of a great impact on the quality, salidroside content get down with time in different varieties of Rhodiola, it may due to chemical changes, The proposed evaluation of Chinese Pharmacopoeia quality medicines should be limited to the validity of Clinical medication to ensure the effectiveness and reliability.The four Chapter , IR identification and quality control in Rhodiola.Comparative analysis of seven species of Rhodiola original drugs powder in on IR and second derivative spectra for the first time, found the characteristics of IR to identify seven different varieties (R. crenulata, R. fastigiata, Rhodiola kirilowii, R.sachalinensis, R.dumulosa et al) IR. Seven varieties of 75% ethanol extract were obviouslier than the original medicinal powder in spectra characteristic information of IR.For the first time , the extraction of petroleum ether, chloroform, ethyl acetate, n-butanol from seven species of Rhodiola drugs is significant than drugs powder in their IR and second derivative spectra. The IR differences can be used as ingredients variety identification reference.For the first time ,using infrared spectroscopy combined with SIMCA Identification Technology in collection of 84 samples of Rhodiola in IR cluster analysis. The preliminary establishment of a seven varieties of indigenous herbal medicine Rhodiola powder mathematical pattern recognition methods, The results show the rate of identification and the refusal were higher than 92%. The pass rate of test samples was 66.7%. SIMCA clustering are applicable to the identification of Rhodiola species. It is a new research ideas for analysis of modern technology in the fields of Rhodiola's identification. |
