Objective: To study the therapeutic effect of Danhong injection on the rats with experimental cerebral ischemia,and investigate the mechanism.Methods:A middle cerebral artery occlusion model of rats were established with autologous blood clots.The rats were divided into 3 groups,including sham operation group,contral group and Danhong Injection treated group.The rats in treated group were injected via tail vein with extracts liquor from Danhong,while rats in both sham operation group and contral group were injected with equal saline,2mL.kg-1,once daily for 5 days. The neurological function was evaluated,and the infarction volume, brain water content, NOS activity,content and activity of t-PA and PAI-1 were measured 1d ,3d and 5d after MCAo.A cerebral ischemia-reperfusion model of mouses were established. Method of grouping and dosage was same as above. The expression of CD41and CD62p in platelet was measured by flow cytometry.Result:1.Compared with control group, NIHSS of therapy was lower at 3d (2.50±0.53 vs 1.80±0.42, p<0.05) and 5d (1.70±0.48 vs 1.10±0.32,p<0.05) after MCAo, Infarction volume was smaller at 5d after MCAo(90.20±16.16 mm3 vs 74.20±9.0 mm3 ,p<0.05). Brain water content was reduced at 3d (82.20±3.46 vs 78.80±1.69,p<0.05) and 5d (83.50±3.63 vs 77.63±1.75, p<0.05) after MCAo,and histopathological changes were alleviated.2. Compared with sham operation group, the activity of total NOS and iNOS of model group were increased(p<0.05), while the activity of total NOS and iNOS of treated group at each time point were lower than in model group(p<0.05).3.Compared with sham operation group, the content and the activity of t-PA from brain tissue and plasma increased at1d,3d and 5d after MCAo.while the content and the activity of t-PA were further increased in treated group,however,compared with control group, but the difference has no statistical significance at each time point.Compared with sham operation group, the content PAI-1 from brain tissue and plasma increased at 1d,3d and 5d after MCAo(p<0.05),while the content of PAI-1 decreased in treated group,however, but the difference has no statistical significance. (P>0.05) at each time point.4.Expression of platelet membrane glycoprotein CD41 and CD62p was up-regulated in experimental mouses early after cerebral ischemia-reperfusion(p<0.05), which maintained for at least 24h. Compared with control group, expression of CD41 was down- regulated in treated group 12h after ischemia-reperfusion(p<0.05),while, and expression of CD62p was down-regulated in treated group 6h to 24h after ischemia-reperfusion(p<0.05).Conclusion:1. Danhong injection could improve neurological function, decrease infarct volume,abate brain edema and histopathological changes, has a significant protective effect on rats with acute cerebral infarction.2. Danhong injection could decrease activity of total NOS and iNOS,diminish the production of NO, therefore had marked anti-oxidative damage effect and provide protective effect against cerebral ischemia.3. Danhong injection dose not be demonstrated to effect content and activity of t-PA and PAI-1 activity in our experiment.4. Platelets was Activated after cerebral ischemia-reperfusion ,while the expressions of CD41 and CD62p in platelets are significantly high.,The expressions of CD41 and CD62p was down regulated in Danhong Injection group. Objective: To investigate the influence of Danhong injection on the therapeutic effect and the changes of TXB2 and 6-Keto-PGFIa in patients with acute cerebral infarction.Methods: One hundred patients with acute cerebral infarction were divided into two groups.The 40 patients in control group were treated with conventional therapy,while the 60 patients in Danhong therapy group were treated with conventional therapy and Danhong Injection.The changes of neurologic function score(NIHSS) ,clinical effect and the content of TXB2 and 6-keto-PGFIa 2 weeks after therapy were compared.Results: NIHSS of control group and therapy group was 8.02±2.13 vs 8.56±1.27 before therapy(p>0.05),while 6.42±1.55 vs 4.36±1.07(p<0.05)2 weeks after therapy. The total clinical effective power of control group and therapy group was 77.5% vs 91.7%(p<0.05).The Serum content of TXB2 from control group and therapy group before therapy was 426.33±224.21 vs 451.52±249.71(p>0.05),while 312.46±176.96 vs 258.59±158.55(p<0.05)2weeks after therapy.The content of TXB2 in the two groups after therapy is decreased compared with that before therapy(p<0.05) . The content of TXB2 of treated group is significant decreased compared with contral group(p<0.05). The Serum content of 6-keto-PGF1 from control group and treated group before therapy was 247.73±106.19 vs 233.90±85.72 ( p>0.05 ) ,while 327.34±134.67 vs 400.94±111.26 ( p<0.05 ) 2weeks after therapy. The content of 6-keto-PGF1 in the two groups after therapy is increased compared with that before therapy(p<0.05) . The content of 6-keto-PGF1 of therapy group after therapy is significant increased compared with control group(p<0.05).Conclusions: Danhong Injection could regulate the balance of TXB2/6-keto-PGF1a, inhibit activity of platelet and improve cerebral blood flow.thereby improve the clinical neurologic function of patients with acute cerebral infaction.
