ObjectiveOn the basis of comprehensive therapy with acupuncture for apoplexy at present,combining modern pharmacological research basement to produce the implantation of collagen and Tetramethylpyrazine,and observing the change of matrix metalloproteinase-9(MMP-9) and tissue inhibitor of metalloproteinase-1(TIMP-1) in cortex tissues of rats with focal cerebral ischemia reperfusion injury after embedding the implantation in acupoints of Baihui and Dazhui.MethodsTotally 96 healthy SD rats were selected and divided randomly into 6 groups,including normal group,sham groups treated for 1d,3d,5d,model groups normal treated for 1d,3d and 5d,groups of embedding medicine in acupoints treated for 1d,3d and 5d,groups of injecting medicine in rats' tail vein treated for 1d,3d and 5d and groups of injecting medicine in acupoints treated for 1d, 3d and 5d,having 6 rats in each group.The model of cerebral artery occlusion(MCAO) and reperfusion was developed by occluding rat' s right middle cerebral artery following suture occlusion method modified by Longa et al.The implantation made up of collagen and Tetramethylpyrazine was implanted into "Baihui"(GV 20) and "Dazhui"(GV14).Every rat in each group wasdecollateddirectly and taken away its cortex tissues in the first 2/3 brain for testing the expression of MMP-9 and TIMP-1 by western blotting.The pictures of western blotting were analyzed with Ge1 base / Ge1 plot in Ge1 Document Analysis System.ResultsResults of western blotting shows:1 Comparing with normal group and sham group,expression of MMP-9 of model groups were higher obviously,and changed all the time,1d group up a little,3d group to the most,and then down by time.The difference was significant(P<0.01).Compared to the same time model groups,expression of MMP-9 in each therapy group was fewer,which was meaningful(P<0.01).The effect of embedding medicine in acupoints was much better than that of injecting medicine inacupoints(P<0.01) and injecting medicine in rat' s tail vein(P<0.01).2 Comparing with normal group and sham group,expression of TIMP-1 of model groups were higher obviously,and changed all the time,1d group up a little,then up by time,5d group to the most. The difference was significant(P<0.01).Compared to model groups, expression of TIMP-1 in each therapy group was more,which was meaningful.The effect of embedding medicine in acupoints was much better than that of injecting medicine in acupoints(P<0.01) and injecting medicine in rat' s tail vein(P<0.01). ConclusionThe expression of MMP-9 in injured cortex tissues turned up obviously and the expression of TIMP-1 in the organization increased also after established the model of rats with focal cerebral ischemia reperfusion.But the balance of MMP-9 and TIMP-1 was broken.TIMP-1 couldn' t stop the injury produced by MMP-9 effectively which broken the minute cerebral vascularities and made the cerebral ischemia reperfusion worse.Embedding medicine in acupoints probably protected rats' brain by turning down the expression of MMP-9 and turning up the expression of TIMP-1 in injured cortex tissues of rats with focal cere- bral ischemia reperfusion injury,then preventing the cerebrovascular endothelial basement membrane from being broken,protecting minute cerebral vascularities,and then lightening the cerebral ischemia reperfusion injury.
