ObjectiveOn the basis of comprehensive therapy with acupuncture for apoplexy at the present time,combining modern pharmacological research to prepare collagen tetramethylpyrazine sustained-elease dose,to research the effects of embedding medicine in Dazhui and Neiguan acupoints on IL-6 and TNF-αexpression in hippocampus in rats with focal cerebral ischemic reperfusion(CIR).MethodsTotally 60 healthy SD rats were selected and divided randomly into 4 groups,The groups included normal group,sham group, model-24h group,model 72h group,model 120h group,embedding medicine in acupoints 24h group,embedding medicine in acupoints 72h group,embedding medicine in acupoints 120h group,with 6 in each group.The model of ischemia reperfusion of cerebral middle artery was established by the modified thread occlusion method,The focal CIR models were established by improved Zea-Longa and then embedding medicine in Dazhui and Neiguan acupoints with collagen tetramethylpyrazine sustained-release dose.each group were decollated directly to investigate the expression of IL-6 and the expression of TNF-αin hippocampus by radioimmunoassay.Results1.Compares between model group and embedding medicine in acupoints group,the symptom of rats' neurologic defect obviously become better(P<0.01).2.After ischemina and reperfusion,the expression of IL-6 and TNF-αin hippocampus serum reinforced(P<0.01).3.The content of IL-6 in hippocampus of model group rats reached peak value in 72h and then dropped gradually,but still maintained at the high level.Compared with model group,the content of IL-6 in hippocampus of embedding medicine in acupoints group was obviously higher(P<0.01).4.The content of TNF-αin hippocampus of model group rats reached peak value in 24h and then dropped gradually.Compared with model group,the content of TNF-αin hippocampus of embedding medicine in acupoints group was obviously lower(P<0.01) and then decreased with the lapse of time.ConclusionEmbedding implantation of collagen and tetramethyrazine in points can decrease the content of TNF-αand rise the content of IL-6 in hippocampus of rat brain after cerebral ischemia reperfusion,cut down neuronal cells damnification by inflammation reaction,and has protection effects on neuronal cells after cerebral ischemia reperfusion in rats,which may be the possible mechanism for on ischemia neurons apoptosis.
