ObjectiveOn the base of therapy cerebral infarction,combining modern phar macological research to prepare collagen tetramethylpyrazine sustaine d- elease dose,to research the effect of embedding medicine in Dazhui and Neiguan acupoints on the infarct volume and its regulating effec t on Fas,heat shock protein-70 and apoptotic cells after focal cerebr al ischemia-reperfusion.MethodsTotally 160 healthy SD rats were selected and divided randomly in to 6 groups,The groups included normal group,pseudo surgery group,m odel-24h group,model 120h group,model 120h group,abdominal cavity i njection 24h group,abdominal cavity injection 72h group,abdominal ca vity injection 120h group,electroacupuncture 24h group,electroacupun cture 72h group,electroacupuncture 120h group,embedding medicine in acupoints 24h group,embedding medicine in acupoints 72h group and emb edding medicine in acupoints 120h group with 10 in each group.The mod el of ischemia reperfusion of cerebral middle artery was established by the modified thread occlusion method,then embedding medicine in Da zhui and Neiguan acupoints with collagen tetramethylpyrazine sustaine d-release dose.4 rats of each group were decollated directly to extra ct unsalted brain tissue with TTC chromating;and other 6 rats were r eperfused with 4%paraformaldehyde after being anaesthetized by 10%c hloral hydrate,and then taken out the brain rapidly.The brain tissue 2mm away from the chiasma was cut frontally,the middle part left,im mersed in hard paraffin.Used immunohistochemistry SABC method to inve stigate the expression of Fas and HSP-70,and the effect of all the a bore index of electro-acupuncture were observed.The entire datum was analyzed by statistics.Results 1.MCAO and reperfusion injury can damage the rats' activity distinctly.The nerve symptoms of abdominal cavity injection group, electro-acupuncture group and embedding medicine in acupoints group were ameliorated obviously;there was a significant difference between model group and each therapy group,and the embinding medicine in acupoints group can improve rats' nerve sympotom remarkably furthermore(compared with abdominal cavity injection group,P<0.01;compared with electro-acupuncture group,P<0.05)2.The expression of Fas heightened in brain tissue after focal cerebral ischemia-reperfusion,and achieved the peak 24h later by the method of immunohistochemistry in model group.There is no expression of FAS in normal group;there are only few positive cells of Fas in pseudo surgery group;the expression of Fas in embedding medicine group is lower than model group significantly(P<0.05,P<0.01;compared with the abdominal cavity injection group and electro-acupuncture group,the differences are also relatively prominent(P<0.05).3.The expression of HSP-70 achieved the peak 24h later and then de clined in brain tissue after focal cerebral ischemia-reperfusion,by t he method of immunohistochemistry in model group,there are only few p ositive cells of HSP-70 in normal group and pseudo surgery group;the expression of HSP-70 in each therapy group are all heightened,and the differences are relatively prominent compared with the same phase-mo del group(P<0.05,P<0.01),and the expression of HSP-70 is increas ing in embedding medicine group especially(compared with the electroacupuncture group,P<0.05,compared with the abdominal cavity injecti on group,P<0.05).4.By utilizing the TUNEL method,the expression of apoptotic positi ve cells is rising and achieved the peak 1 days later ischemia-reperf usion,then declined.There are only a few apoptotic cells in normal g roup and pseudo surgery group;the expression of apoptosis in embeddin g medicine group,electro-acupuncture group,abdominal cavity injectio n group are all lower than the model group(P<0.05,P<0.01),especia lly the embedding medicine group(P<0.05).ConclusionThe expression of Fas increased and apoptosis celles in ischemica l brain tissue after focal cerebral ischemia reperfusion in rats,whic h results in the neuron apoptosis and enlarges brain infarct volume,a ggravated ischemia reperfusion injury.Embedding medicine in acupoints can decrease the expression of Fas and apoptosis celles and increas e the expression of HSP-70,so it can synthesize protective neuron a nd depress apoptosis,consequently,exert the effect of neuron protect ion and bring into play the effect of cerebral protection.
