"Gugongguogen" is the root of Rose odorata Sweet var.gigantea(Coll.et Hemsl.) Rehd.et Wils which distributes in Yunnan Province,the south of China.The experiments of pharmacological activity indicated that the chemical constituents from the root showed strong anti-diarrhea activity.By now,no data about its chemical composition has been reported.In this dissertation,the dried root of Rose odorata Sweet vargigantea(Coll.et Hemsl.) Rehd.et Wils was extracted with 60%ethanol,and 15 compounds were isolated using various chromatographic techniques,such as silica CC,ODS CC,from the EtOAc layer.Their structures were elucidated on basis of physico-chemical characters,spectroscopic analysis(IR, MS,1H-NMR,13C-NMR and 2D-NMR).These compounds areβ-sitosterol(1),betulinic acid (2),2α,3β,19β-trihydroxyurs-12-en- 28-oic acid(3),euscaphic acid(4),2α,3α,19α, 23-tetrahydroxyurs- 12-en-28-oic acid(5),2α,3β,19α-trihydroxyurs-12-en-28-oic acid(6), kaji-ichigoside F1(7),rosamultin(8),gallic acid(9),catechin(10),(-)-epicatecbin(11),2α, 3α,19α,23-tetrahydroxy-urs-12-en-28-oic acid-3-O-β-D-glucopyranosyl ester(12),ilexoside B(13),(4α,8)-(-)-fisetinidol-(-)-epicatechins(14),procyanidin B3(15).Compounds 13,14 were isolated from Rosa genus,for the first time.Compounds 1,2,3,4,5,6,7,8,9,10,11, 12,15 were isolated from this plant for the first time.Compound 13 showed stronger activity against NO releasing in RAW 264.7,and at a lower concentration,no cytotoxic activity was observed in bioassay.And we also studied the technological parameters in the purification process of total tannins from "Gugongguogen" by macroporous resins for the first time.Using adsorptive quantity as the guideline,the adsorptive capabilities and static eluation ratio of eight kinds of resins were compared,and the optimal macroporous resins were selected.The influence factors of purification of total tannins from "Gugongguogen" with the optimal macroporous resin were tested and screened by the adsorption and elution ratio of total tannins.In conclusion,this research has provided rational and experimental basis for further development of this drug.
