Objective:To investigate the influence of Gypenosides(GP)and water extraction of Polygonatum Odoratum(PO) on the proliferation of 3T3-L1 adipocytes and insulin resistance.Method:1.Normal male Wistar rats were randomly divided into eight groups:normal group,control group,GP groups of low, moderate and high dosage,PO groups of low, moderate and high dosage. After 10 days of orally treatment,rats were taken blood.2. 3T3-L1 preadipocytes were cultured,the proliferation of 3T3-LI preadipocytes and 3T3-L1 adipocytes were detected by MTT method.Insulin resistant 3T3-L1 adipocytes cell model was induced by dexamethasone and insulin.The change of glucose concentration in cell culture was determined after GP and PO treatment.Result:1. MTT method showed that the absorbance at 510nm of 3T3-L1 adipocytes was decreased after treated by GP for 24h(P<0.05).GP significantly increased glucose consumption in IR 3T3-L1 adipocytes culture in a concentrateion-dependent manner(P<0.05).2. PO groups didn't increase glucose consumption in IR 3T3-L1 adipocy- tes culture. PO group of high dosage had toxic effect on cells.Conclution:1. GP can inhibit the proliferation of 3T3-L1 adipocytes,and also,significantly improve insulin resistance induced by dexamethasone. Promoting the glucose consumption of induced insulin resistance adi- pocytes may be one of possible hypoglycemic mechanisms of GP.2.PO probably can't promote the glucose consumption of induced insulin resistance adipocytes to improve insuline resistance.
