Wenyanghuoxue Regulation Of Traditional Chinese Medicine Within The Coronary Heart Disease Platelet Receptor - Ligand, Signal Transduction Mechanism Of Action Study

Objective: This study was designed to observe the effect of Chinese compound prescription with the function of warming yang and activating blood to the yang deficiency and blood stasis symptom of coronary heart disease rats platelet receptor-ligand binding and signal transduction through animal experiments observation.Thereby revealing the mechanism of the compound to preventing and treating coronary heart disease.Methods: Using the syndrome combination modeling methods to establish models of the yang deficiency and blood stasis symptom of coronary heart disease rats. Using advanced physical and chemical examination methods such as color Doppler,ECG computer software PCLAB biological signal acquisition and processing system and enzyme-linked immunity method,biochemistry,electron microscope and other modern detection techniques,to analyse and verify the validation of animal models. And observing the effect of platelet receptor-ligand binding,signal transduction on platelet activation from multiple levels,and cellular and molecular level. Understanding the correlation of the signal transduction mechanism of platelet activation and the yang deficiency and blood stasis symptom of coronary heart disease. Providing more accurate objective indicators which revealing the essence of the yang deficiency and blood stasis symptom and the mechanism of Chinese medicine to platelet receptor-ligand binding and signal transduction of coronary heart disease patients.Results: The rat model successfully simulated the main pathological changes of clinical coronary heart disease cardiac dysfunction,i.e. blood fats ascension and the cardiac muscle lacks the blood. The rat model generated the changes as follows,platelet ligand vWF ascended,the amount of platelet receptor membrane glycoprotein GPⅡb/Ⅲa molecule increased,arachidonic acid metabolites TXA2 increased and PGI2 decreased,α-granule membrane protein GMP-140 increased,the rate of platelet aggregation increased.Conclusion: Chinese compound prescription with the function of warming yang and activating blood could block the combination of platelet receptor–ligand,could regulate the Cascade response of signal transduction,could reverse the changes of rats model platelet function and morphology, could reduce injury in rats model, and could prove that the rats model possess sickness nature of he yang deficiency and blood stasis symptom.
