There is frequent relationship between changes of blood flow and the function of gastrointestinal tract as well as the cause,development and recovery of Gastrointestinal tract pathological changes.For instance,gastrointestinal mucosal ischemia leads to the degradation of gastrointestinal mucosal function and the disorder of epithelial cell metabolism,therefore results in gastrointestinal hemorrhage and necrosis of the mucosa.While Gastrointestinal mucosal hypoperfusion ischemic is one of the main reasons for stress-induced gastrointestinal mucosal injury.Gastric mucosal microcirculation is the pathophysiological process of Stress ulcer.Various factors are attributed to the disorder of gastric mucosal microcirculation,which includes:Intravascular influences,such as the activation of leukocyte,platelet,and the changes of hemorheology;Vascular wall-based,such as vasoactive substances like NO and ET; Extravascular-based,such as glutamate release,Ca2+influx and etc..Studies on how electroacupuncture treatment affects above factors have been conducted,however,it is Rare that how electroacupuncture will affect cell adhesion molecules,particularly the E-selectin. The significance of E-selectin in ischemia-reperfusion injury is it meditates leukocyte rolling in the areas of inflammations and injured tissues,and activates endothelial cells' adhesions with neutrophils.In order to investigate the mechanism of how electroacupuncture affects the changes of gastric blood flow thoroughly,this study firstly researchs the effects of electroacupuncture on the changes of gastric blood flow and the variations of gastric functions,on the base of this it further discusses electroacupuncture's effects on E-selectin.This study includes two sections: Literature review and experimental study.The literature review includes two summaries: Doppler imaging diagnostic techniques and its medical applications;E-selectin and Inflammatory injury.The experimental study has two sections:1.MethodAfter surgery,healthy adult SD rats were induced to gastric ischemia-reperfusion—Single-organ ischemia-reperfusion models(SOIRM).The models were divided into two groups:The electroacupuncture-treated group(Studied group)and the non-electroacupuncture-treated group(Contrasted group).The laser doppler perfusion imager(LDPI)was used to observe the effects of electroacupuncture on the changes of gastric blood flow of the SOIRM.Multilead physiological recordinginstrument was applied to record the effects of electroacupuncture on the changes of bioelectricity(Fast-wave)of SOIRM.And immune staining method was used to analyse the effects of electroacupuncture on the changes of E-selectin on SOIRM.2.Results2.1 The effects of electroacupuncture treatment on the changes of gastric blood flow of the SOIRMThe trends of the changes of gastric blood flow on both the electroacupuncture-treated group and the contrasted group observed by LDPI indicates no difference Obviously.That is when ligating right gastric artery,the blood flow decreased,while when loosening the ligation, the blood flow increased subsequently.The difference is that the extent of ischemia of the studied group is lower than that of the contrasted group.The reperfusion indicates that the degree of recovery of the blood flow of the studied group is greater than that of the contrasted group.Therefore,this shows that electroacupuncture treatment has affects on the improvement of blood circulation particularly.The statistical difference between the results of the analysis is:During the ligation,the decrease volume of the blood flow of the studied group is lower than that of the contrasted group.At 25 min,the studied group and the contrasted group has significant statistical difference(i.e.t=2.135,P<0.05);in the loosening stage,the increase volume of the blood flow of the studied group is greater than that of the contrasted group.2.2 The effects on the changes of gastric bioelectricity of SOIRM by Electroacupuncturing Zusanli Point in the ratsThe amplitude of gastric electricity of the contrasted group has an decreasing increasing tendency on both circumstances(Litigation and loosening),and there is no significant statistical difference(P>0.05).The studied group shows that when litigation is induced,the amplitude of gastric electricity has significant increase compared with the circumstance of loosening(P<0.05).An initial increasing tendency is detected and then it decreased.The cluster numbers of fast wave of gastric electricity in the contrasted group has significant difference under the litigation stage compared with the normal state(P<0.05),the change of the fast wave clusters increased after the first drop.The cluster numbers of fast wave of gastric electricity in the studied group has a decreasing tendency.There is significant statistical difference between the loosening state and the normal state(P<0.05).2.3 The effects of electroacupuncturing the Zusanli Point(ST36)in the rats on E-selectin expression of vascular endothelial cells of StomachThe results of E-selectin expression indicate that in the contrasted group,there are small amount of positive staining reactions which are microvessels and capillary vessels(Brown colour),small vessels and great vessels has the negative reactions.The studied group shows it has small amount of positive staining reactions but the amount is less than that of the contrasted group.The areas of E-selectin expression of the gastric tissues of the contrasted group have an increasing tendency Significantly(P<0.01).After the treatment of electroacupunctue,the areas of E-selectin expression of the gastric tissues of the studied group have decreased compared with that of the contrasted group.However,it does not have significant statistical difference.There is a significant statistical difference between the E-selectin expression between the studied group and the contrasted group(P<0.01). 3.conclusionThe laser Doppler perfusion imaging showed that the gastric wall blood flow gastric ischemia is reduced than the controlled group significantly,the gastric fast wave of the EGG amplitude and frequency are declining than the normal state,the E-selectin expression of endothelial cells in the gastric wall microvascular and capillary vessels increased than the normal groupsignificantly;the blood flow of the gastric wall reperfusion is increased than the previous significantly,the gastric fast wave of the EGG amplitude and frequency are still a downward trend compared with the normal state,the E-selectin expression of endothelial cells in the gastric wall microvascular and capillary vessels is increased than the normal group significant.After electroacupunctured the Zusanli point,the gastric ischemia and reperfusion of blood flow in two states could improved,but there is no significant difference compared with the control group;the the impact of electroacupuncture to the fast wave amplitude is increased further to lower,the change of frequency is reduced,the electrical activity of the stomach is the role of inhibition in general;the E-selectin expression of endothelial cells in the microvascular and capillary vessels in the electro-acupunctured group is on a decreasing trend,but no statistical difference compared with the control group.
