Dan-shen,the dried roots of Salvia miltiorrhriza Bunge,is one of the most popular traditional Chinese medicines,the tanshinones which was separated from Dan-shen has been widely used by for promoting blood stasis,cleaning away heat,relieve vexation, nourishing blood and tranquilizing the mind and cooling blood to relieve carbuncles,such as TanshinoneⅡ_A,TanshinoneⅠand Cryptotanshinone.This research has studied the extraction and separation of total tanshinones from Dan-shen and their Structure modification were investigated.1.Tanshinone extraction and separation process is divided into three main steps.One step:To study the effects of different extraction methods(Heating and refluxing extraction, Ultrasonic extraction and Smashing extraction) on the content of TanshinoneⅡ_A and Salviandic acid B from Dan-shen,the optimum processing were determined,The smashing extraction method was first applied during the extraction of tanshinones from Dan-shen. Two step:the method combined The smashing extraction and Ultrasonic extraction is first applied during the extraction of tanshinones and significantly increases the extraction rate of tanshinones from Dan-shen.Three step:finally tanshinone was extracted from the root of Salvia miltiorrhiza Bunge separated by column chromatography.2.Aldehyde reaction,bromination reaction and basic hydrolysis reaction of TanshinoneⅡ_A and cryptotanshinone were also discussed in this work,The reactions' mechanism were also proposed.①The reaction of tanshinoneⅡ_A with aldehydes could be carried out in soft condition by p-methyl-benzene sufonic acid catalyzing and ten 2,2'-(substituted methylene) bis(1,6, 6-trimethyl-6,7,8,9-tetrahydrophenanthro[1,2-b]furan-10,11dione) derivatives were obtained in excellent yield.②TanshinoneⅡ_A can occur with liquid bromine electrophilic substitution reaction, Product is 2-bromo tanshinoneⅡ_A,and the Bromination product of cryptotanshinone is complex.③Cryptotanshinone can be in open-loop in 2mol/L of NaOH solution,then,tanshinoneⅡ_A that its role in the alkali can be irreversible open-loop and its structure was damaged can't be open-loop in dilute alkali solution. The subject is the structural transformation on the D ring of tanshinone,and lay the foundation for further revealing the physiological activity of tanshinone relationship with the D ring,and at the same time compared the chemical properties of cryptotanshinone with tanshinoneⅡ_A.then get a total of 12 derivatives.
