Found The Wind Expectorant On Angiotensin Ii-induced Lox-1 Mrna And Of Et-1 Mrna Expression In Endothelial Cells

Purpose:to observe the effects of expelling wind to eliminate phlegm on the expression of human umbilical vein endothelial cells(HUVECs)LOX-1 mRNA and ET-1 mRNA in vitro induced by angiotensin II.Material and method:The serum containing metabolic ingredients of Huoxin Decoction was taken from rabbits. HUVECs2-3 were cultured in vitro by collagenase digestive method and HUVECs were randomly divided into five groups:normal control group, Ang II group, low concentration drug serum and Ang II group, middle concentration drug serum and Ang II group, high concentration drug serum and Ang II group. LOX-1 mRNA and ET-1 mRNA were identified with RT-PCR method.Results:1. The expression of LOX-1 mRNA and ET-1 mRNA could be increased obviously by the injury of endothelial cells induced by Ang II.2. Huoxin Decoction could inhibit the injury of endothelial cells and decrease the expression of LOX-1 mRNA and ET-1 mRNA.3. The expression of LOX-1 mRNA and ET-1 mRNA of the 3 drug serum groups was sharply lower than that of Ang II group (P<0.05); the expression of LOX-1 mRNA and ET-1 mRNA of the middle and high concentration drug serum group was significantly higher than that of the low concentration drug serum group and there was no significant differences of it between the middle and high concentration drug serum group.Conclusion:1. Huoxin Decoction of different concentration could inhibit the injury of HUVECs induced by Angâ…¡and decrease the expression of LOX-1 mRNA and ET-1 mRNA. There was dose-effect relationship in this phenomenon.2. Huoxin Decoction could inhibit the injury of HUVECs induced by Angiotensinâ…¡to protect vascular endothelial in order to control coronary atherosclerosis.
