Purpose:This study, application of Molecular biology and biochemical methods, by Angllon cultured vascular endothelial cells LOX-1mRNA, NF-KBmRNA, iNOSmRNA and ET-1mRNA activation and the impact of Chinese mecicine-intervention, the use of wind removing phlegm Chinese search studies on the regulation of Herbs to explore traditional Chinese medicine Angâ…ˇon human umbilical vein endoth-elial cells induced by LOX-1mRNA, NF-KBmRNA, iNOSmRNA and the expression of ET-1mRNA. Thus for research and development new drug of TCM of treatment atherosclerosis provide scientific theory basis.Material and method:1. The 20 healthy White rabbits is randomly divided into four groups:low-dose group, medium-dose group, high-dose group and blank control group. The first three dose groups separately Huoxin Decoction intragastric, white space, such as volume control group be distilled water after intragastric and successful model for blood treatment of abdominal aorta, rabbits Huoxin Decoction Chinese herbal compound containing serum.2. With enzyme digestion method in vitro generation of HUVECs2-3 used in the experiment.3. After an experimental medicine interventions will be randomly divided into 5 groups:rormal control group (blank serum); Angâ…ˇgroup (0.5ÎĽmol/L Angâ…ˇ);Ang-â…ˇand low serum concentration of traditional Chinese medicine (0.5ÎĽmol/L Angâ…ˇ+2.5% TCM serum); Angâ…ˇand in the serum concentration of traditional Chinese medicine (0.5ÎĽmol/L Angâ…ˇ+5% TCM serum); Angâ…ˇand high concentration of Chinese serum (0.5ÎĽmol/L Angâ…ˇ+10% TCM serum).4. Adopted by RT-PCR method for determing expression levels of human umbilical vein endothelial cells LOX-1mRNA, NF-KBmRNA, iNOSmRNA and ET-1mRNA.Results: 1. Angâ…ˇcan lead to human umbilical vein endothelial cells (HUVECs) injury, the LOX-1mRNA, NF-KBmRNA, iNOSmRNA and ET-1mRNA expression was significantly increased;2.Huoxin decoction can inhibit Angâ…ˇinduced endothelial cell LOX-1mRNA, NF-KB mRNA, iNOSmRNA and ET-1mRNA expression;3. Inhibition of different concentrations of Chinese serum LOX-lmRNA, NF-KBmRNA, iNOSmRNA and the effect of ET-1mRNA expression differences. Among them,the middle and high concentration drug serum group inhibition of the expression of LOX-1mRNA, NF-KBmRNA, iNOSmRNA and ET-1mRNA more obvious than that of the low concentration drug serum group, and there was no significant differences of it between the middle and high concentration drug serum group,but the expression in the middle concentration drug serum group is reduced evidently than that of the low concentration drug serum group and there was statistical differences.Conclusion:1. Angâ…ˇcan lead to endothelial cells injury, the LOX-1mRNA, NF-KBmRNA, iNOSmRNA and ET-1mRNA expression was significantly increased;2.Search the wind and Eliminating Phlegm Huoxin Decoction can inhibit endothelial cell LOX-1mRNA, NF-KBmRNA, iNOSmRNA and ET-1mRNA expression,which play to its anti-atherosclerotic effect;3. Huoxin decoction containing serum group inhibit LOX-1mRNA, NF-KBmRNA, iNOSmRNA and the effect of ET-1mRNA expression levels with the Huoxin decoction concentration into a certain of the dose-effect relationship. Compared to the low concentration drug serum group, the middle and high concentrations of serum groups inhibite LOX-1mRNA,NF-KBmRNA, iNOSmRNA and the effect of ET-1mRNA expression more obvious, and no significant difference between the two groups, while the expression in the middle concentration drug serum group is reduced evidently than that of the low concentration drug serum group and there was statistical differences.
