Purpose: To investigate the influences of Salvianolic acid B on the hemodynamics and heart function on isolated normal rat heart and the protective effects of Salvianolic acid B on isolated myocardial ischemia/reperfusion injury in rats.Method: The Langendroff model were prepared and the effects of Salvianolic acid B on left ventricular systolic pressure(LVSP), left ventricular end-diastolic pressure (LVEDP), intraventricular pressure maximum upstroke velocity(+dp/dtmax), intraventricular pressure maximum decrease velocity(-dp/dtmax) and heart rate(HR) on normal rat heart were observed. The ischemia/reperfusion injury model was set up by isolated working heart perfusion technique. Coronary outflow were collected. The effects of Salvianolic acid B on AST, LDH, SOD, and MDA were observed.Result: Compared with normal control group, the LVSP, +dp/dtmax, -dp/dtmax as well as HR were decreased by the perfusion of Salvianolic acid B. The results showed that Salvianolic acid B decreased the release of AST and LDH in coronary outflow at different time point and exhibited protective effects on myocardial ischemia/reperfusion infury. Salvianolic acid B increased SOD activity and decrease MDA level in myocardium. Conclusion: Salvianolic acid B enhances the left ventricular systolic function of the isolated rat heart. Salvianolic acid B shows apparent protective effect on myocardial ischemia/reperfusion injury.
