Objective: In this experiment, through observing the changes of pressure overload heart failure model and the effects of cardiac function by aerobic exercise intervention on the early heart failure rats, to explore what is the myocardial adaptation mechanism of aerobic exercise intervention of the early heart failure rats.Methods: 90 male Wistar ratswere randomly divided into two groups, 60 by abdominal aortic coartation operation made of heart failure(HF) model for operation group, 30 only separated abdominal aorta not be coarctated, other processing was the same as the operation group. 8 weeks after operation, the operation group were randomly divided two groups,30 operation without training group(Rest),30 operation training group(Exer),and the sham sham group is the sham operation without training(Sham).Group Exer performed treadmill training, exercise intensity of 20 meters / minute, 1 hour a day, 5 days a week of training, and totally 8 weeks training. After 8 weeks of training, detecting the rats LVID,IVS,LVPW,EF,FS and SV by Visual Sonics Vevo®2100; detecting the rats LVSP、 LVEDP、HR and ±dp/dt max values by Lab Chart7;and determining the state3 respiration, state 4 respiration and respiratory control ratio(RCR) of myocardial mitochondrial by O2 K.The protein expression of myocardial UCP2,HIF-1alpha /beta were determined via western blotting.Result: 1. The changes of cardiac structure after HF and the effect of aerobic exercise on it1) Compared with group Sham, the rats of group Rest cardiac structure indexes LVID; s, LVID; d, IVS; d, LVPW; s, LVPW;d were significantly increased.2) Compared with group Rest, the rats of group Exer cardiac structure indexes LVID; s, LVID; d, IVS; d, LVPW; s, LVPW; d were significantly reduced; Compared with group Sham, there were no significant difference.2. The changes of cardiac function after HF and the effect of aerobic exercise on it1) Compared with group Sham, the rats of group Rest cardiac function indexes EF, FS and SV values were significantly reduced and the value of LV Vol;s was significantly increased, cardiac hemodynamic indexes LVSP、 LVEDP、HR were significantly increased and ±dp/dt max values were significantly reduced.2) Compared with group Rest, the rats of group Exer cardiac function indexes EF, FS and SV values were significantly increased and the value of LV Vol;s was significantly reduced, cardiache modynamic indexes LVSP、 LVEDP、HR were significantly reduced and ±dp/dt max values were significantly increased.Compared with group Sham, there were no significant difference.3. The changes of cardiac mitochondrial function after HF and the effect of aerobic exercise on it1) Compared with group Sham, the rats of group Rest cardiac mitochondrial respiratory function indexes of state 3 respiration, RCR values decreased significantly but the value of state 4 respiration was significantly increased.2) Compared with group Rest, the rats of group Exer cardiac mitochondrial respiratory function indexes of state 3 respiration, RCR values increased significantly and the value of state 4 respiration was significantly reduced. Compared with group Sham, there were no significant difference.3) Compared with group Sham, the rats of group Rest complex I activity decreased significantly. Compared with group Rest, the rats of group Exer complex I activity was significantly increased.there were no significant difference in the activity of complex Ⅱ/Ⅳ among three groups.4. The changes of the expression of UCP2,HIF-1α/β protein after HF the effect of aerobic exercise on it1) Compared with group Sham,in the rats of group Rest,the expression of UCP2 protein increased significantly; the expression of HIF-1α protein decreased significantly and the expression of HIF-1β protein was significantly increased.2) Compared with group Rest, in the rats of group Exer, the expression of UCP2 protein decreased significantly; the expression of HIF-1α protein and HIF-1β protein were no significant difference. Compared with group Sham, in the rats of group Exer, the expression of UCP2 protein was no significant difference; the expression of HIF-1α protein decreased significantly and the expression of HIF-1β protein was significantly increased.
