There is increasing interest in the saidies on the prevention and treatment of postmenopausal osteoporosis (PMOP) all over the world. In order to investigate this issue further we carried out three kinds of experiments as follows.PART I(-) Effect of Short-term-Calcium supply on Bone Quality and Bone Metabolism in Ovariectomized RatsOBJECTIVE To discuss the effect of short-tenn-calcium supply on bone quality and bone metabolism in ovariectomized rats. METHOD 25 female SD rats were divided into 3 groups: A group: SHAM group; B group: OVX group: C group: OVX+Calcium ( Ca) supply group. After 5 weeks, the rats were killed and BMJ) BMC~ biochemical markers and biomechanical parameters were assayed. RESULT (1) BMD of A group was higher than B group by 30/o(P<0.05); B group and C group had almost the same amount of BMID. (2) BMC of A group was higher Iban B group by 12%(P<0.05); BMC and ash weight of C group were more than B group, respectively by 4% and 12% (both P<0.OSX, (3) Concentrations of serum ALP~ Ca and P of A group were significantly higher than B group (respectively 40%~ 112% and 73%); Those of C group were significantly higher than B group (respectively by 75%~ 164% and 57%). CONCLUSION (1) Short-tenn-Ca supply could improve the bone mass and bone strength in OVX rats, but not so significantly as long-term-Ca supply. (2) Ca supply had significant effect on bone metabolism. It maybe stimulate bone formation.(T2i) Effects of Low Frequency Magnetic Fields (ELFMF) on Bone Quality and BoneMetabolism in Ovariectontized RatsOBJECTIVE To investigate mechanisms of prevention and treaUnent of osteoporosis using lowfrequency magnetic fields. METHOD 34 female SD rats were divided into 4 groups, ASHAM group; B OVX group; C OVX+ magnetic field 30' /day; D OVX+magnetic field60' /day. RESULT (1) BMD of D group was higher than B group and C group (P (o.os. However, there was no significant difference in BMD between C group and B group. (2)Dry weight of femurs ash weight of femur and bone mineral content in D group are much higher than B group (P (0.05). Bone mineral content of C group was much higher than B group (P (0.05). (3) Concentrations of serum ALP. Ca and P of C group and D group were all significantly higher than B group( P (0.05). CONCLUSION (1 )Low frequency magnetic fields.5could significantly improve the BMD and bone strength. (2) Low frequency magnetic fields alsocould significantly stimulate bone formation.PART II3MATERIAL Qsteoblast-like cell OS-732; METHOD Cell counting~ Determination of MTL Alkaline phosplate (ALP) stain~ Investigation of ALP production.(- DThe Proliferative and Differentiative Effect of Alendrotiate ( ALN ) on Osteoblast-like cell OS-732RESULT (1 )There days after ALN was added, the cell number of lO~ group was more than control group by ~2.6%. Six days after ALN was added, the cell numbers of l0~% 107 and lO~ group were still more than control group (respectively by 24.7%~ 24.7% and 31.00/.) The determination of MIT had similar results. The cell number of I 0~ group was significantly less thancontrolgroup. (2) FourdaysafterALNwasadded, l0~'.. lff7and l(Y8grouphadmuch..large and regular cells. However, control group had few.. small and uregular cells. Cells in l0~ group grew better than other groups. (3) The result of ALP stain and the investigation of ALP level showed concentrations of ALP in 1 0%nd 10~ group were significantly higher than control group.
