| The paper studied genetic diversity in Liaoning natural Korean pine populations with ISSR method. Thirteen ISSR-PCR primers that had vivid amplification bands and abundant polymorphism had been screened. DNA sample were chosen form different regions of Linging5 natural Korean pine populations. DNA was extracted form the Korean pine needles by improved CTAB method that was high for purity of DNA and met the needs of a large number of PCR amplification. The purity of DNA that was best stable repeatable Korean pine ISSR-PCR reaction systems.In this paper the results of the genetic diversity suggested that were able to116 loci in total, there were 90 polymorphic loci and the percentage of polymorphic loci is 77.59%; Shannon's index is 0.4461, and Nei's index is0.3005.Korean pine populations have a high genetic diversity. Genetic diversity between populations possessed 19.81% and genetic diversity among populations possesed73.09%.We can conclude that genetic diversity mostly exit in populations. According to 5 natural Korean pine populations, population genetic distances of its UPGMA cluster. It can be concluded that that Feng cheng sha zi gang, Kuan dian lin chuan, Kuan dian bai shi la zi together in one group, the Ben xi ba li dian and Fu shun xin bin gang shan come together into one group, the classification of the results and actual location are related. |
