Hericium erinaceus is nutritious and delicious,and most of its researches had focused on deep processing in recent years.There are few studies on the identification and classification of fungi.In the past,the identification and classification of Hericium erinaceus varieties were mostly classified by morphologi cal classification,and the classification of Hericium erinaceus could not be fu ndamentally accurate.Studies had shown that species could be distincted by DNA bar code technology,and even new species can be found and breeding efficiency will be greatly improved.In this paper,molecular biology methods were used to study.And the ?t ubulin gene was selected as the target gene of the bar code sequence.Based on the full length of the ?tubulin gene sequence of Hericium erinaceus in th e NCBI,a pair of primers for the amplification of the target gene were design ed: BetuF1:ATGCGTGAAATCGTCCACC;BetuR2:GAAGACGGGGGAAAGGAAC.The above primers were used to identify the 19 varieties of Hericium erinaceus.Sequencing sequences were analyzed by BLAST,Clustal,MEGA,Bio Edi t and other software,and the polymorphic loci were obtained.The specific primers were designed by analyzing the polymorphic loci.The results showed that three unique barcode of Hericium erinaceus RT9 were successfully obtained.DNA barcode technology is simple,fast,accurate in species identificationand classification,but the technology in the field of research on fungi startedlate,no target genes have been identified so far.Therefore,I hope the resultsof this experiment can not only provide help for further research related aspects but also raise the concern of this technology from the relevant departments and researchers in this kind of research,and then promote the development offungi DNA bar code technology in related fields.
