| With the improvement of living level, mutton becomes peoples' favorite meat owing to its low fat, low cholesterol, high protein and pollution-free. The fat (Intramuscular fat, IMF) content is important to evaluate the quality of muscles. H-FABP is the major gene which influences the precipitation of IMF. The H-FABP polymorphism makes the cells uptake the fatty acids and has important biological functions in the form of meat. Therefore it has a somescitific and practical value for the molecular genetics breeding and marker-assisted selection of the animals when the H-FABP is studied with the production traits as a canditate gene.Direct sequencing was used to find the SNPs at the H-FABP gene in 98 ChongMing white goats,100 ChongMing hybridized goats,91 XuHuai goats and 65 GuanZhong milchgoats from ChongMing Island. PCR-RFLPs were established to differentiate the SNPs; Soxhlet extractor was used to identify the fat content; SPSS was used to analyze the genetic relationship between polymorphisms of H-FABP gene and the association with intramuscular fat content.There were one mutation (A1017G) and one vacancy (G999-) at this gene locus. A1017G makes up a new Haeâ…¡restriction enzyme cutting site, G999- makes up a new Sauâ… restriction enzyme cutting site. A1017G can be analysised by Haeâ…¡-RFLP, and there were BB, Bb, bb three genotypes. The BB was the predominant genotype. A chi-square analysis suggested that the allele frequencies and genotype frequencies of H-FABP gene were not in Hardy-Weinberg equilibrium in the ChongMing white goats, the ChongMing hybridized goats and the XuHuai goats, and the PIC values of these goats were all low, PIC<0.25; There was only BB genotype in the GuanZhong milchgoats, and there was no polymorphisms. G999- can be analysised by Sau I-RFLP, and there were DD, Dd, dd three genotypes. The DD was the predominant genotype. A chi-square analysis suggested that the allele frequencies and genotype frequencies of H-FABP gene were not in Hardy-Weinberg equilibrium in the ChongMing white goats, and the PIC value was low, PIC<0.25; There was only DD genotype in the ChongMing hybridized goats, the XuHuai goats and the GuanZhong milchgoats, and there were no polymorphisms, the skewness is serious. H-FABP gene increased intramuscular in the order of Bb>bb>BB, dd>Dd>DD. It meant that the fragment of H-FABP had polymorphisms, which could be used as a candidate gene associated gene with phenotypic traits like intramuscular fat content in different goat populations. |
