Polymorphism Of H-FABP Gene And Its Association Genetic Effects On Economic Traits In Wei Pig And Wannan Black Pig

In this study, modern molecular maker techniques were used to analyse the polymorphism of5’-upstream region and the second intron, then clone sequencing analyse them in two Anhui superior indigenous pig breeds (Wei Pig, Wannan Black Pig) heart fatty acid-binding protein (H-FABP) gene, then association with carcass traits and meat quality. In order to provide certain basis for protection and exploitation in this two Anhui indigenous pig breeds, and provide the theoretical foundation for commercial strains breeding.The results show that:(1) There were polymorphism in site of the HinfⅠ, in Wei Pig and Wannan Black Pig5’-upstream region, and the frequency of allele H was0.6545and0.6223, both of them show moderate (PIC=0.3575,0.1307), respectively. In the second intron, site of the Hae Ⅲ also existed, allele D frequency was0.6301and0.9239, showing moderate polymorphism (PIC=0.3575). and low polymorphism (PIC=0.1307), respectively. We have just detected two genotypes, DD and DD, in Wannan Black Pig. While no polymorphisms were detected in the site of Msp Ⅰ, the genotype s were AA. The polymorphism in site of Hinf Ⅰ*also existed in two indigenous pig breeds, and the frequency of allele B was0.7195and0.7826, both of them show moderate (PIC=0.3222,0.2824), respectively. Chi-square test showed that, the gene frequencies and genotype frequencies have reached the Hardy-Weinberg equilibrium (P>0.05) in Test group. except the HinfⅠ polymorphic site of the H-FABP gene5’-upstream region in Wannan Black Pig.(2) Cloning and sequencing analysis on polymorphic fragments of three PCR-RFLP, in Wei Pig and Wannan Black Pig H-FABP gene, showed that HinfⅠ-RFLP is due to T→C mutation in1324bp, HaeⅢ-RFLP is due to C→G mutation in1811bp, Hinf Ⅰ*-RFLP is due to C→T mutation in1970bp.(3) The correlation analysis of polymorphic site of H-FABP gene with carcass traits is showed that, in site of the HaeⅢ, carcass lean percent of Dd genotype, in Wei Pig, showed significantly higher than dd genotype (P<0.05), and showed the trend with Dd>DD>dd. In site of the Hinf Ⅰ*, slaughter rate of Bb genotype, in Wannan Black Pig, showed significantly higher than bb genotype (P<0.05), and showed the trend with Bb>BB>bb. Besides, genotypes of other site did not reach the level of significant difference (P>0.05) in the other index of carcass traits. (4) The correlation analysis of polymorphic site of H-FABP gene with meat quality is showed that, in site of the Hinf Ⅰ, the pork color score of HH genotype, in Wei Pig, showed significantly higer than hh genotype(P<0.05), and showed the trend with HH>Hh>hh. The a*value of Hh genotype, in Wannan Black, showed significantly higer than hh genotype(P<0.05), and showed the trend with Hh>HH>hh. The marbling score in the HH genotype Hh significant exceed genotype(P<0.05)in Wei Pig and Wannan Black Pig. In site of the Hae Ⅲ, The L*value of Dd and dd genotype, in Wei Pig, showed extremely exceed DD genotype(P<0.01), showed the trend with Dd>dd>DD. This testing result is close to the Wannan Black Pig detection result. The muscle water in the DD genotype extremely exceed Dd and dd genotype (P<0.01)in Wei Pig, showed the trend with DD>Dd>dd. The DD and Dd genotype showed significant influence in pork color score (P<0.05), showed the trend with DD>Dd. In site of the Hinf Ⅰ*, The drip lost of Bb genotype, in Wannan Black, showed significantly higer than bb genotype(P<0.05), showed the trend with Bb>BB>bb.(5) The correlation analysis of polymorphic site of H-FABP gene respectively to the intramuscular fat is as follows: the effecting tread of detecting genotype to the fat content in the muscle is HH>Hh>hh, dd>Dd>DD. Among that, the content of IMF in the HH genotype extremely exceed Hh genotype(P<0.05), but it extremely significantly exceed hh genotype(P<0.01), Hh genotype extremely exceed hh genotype(P<0.05), dd genotype extremely exceed Dd genotype(P<0.05), but it extremely significantly exceed DD genotype(P<0.01)except for Wannan Black pig, Dd genotype extremely exceed DD genotype(P<0.05). There was no significant difference among the there genotypes in IMF content in Wei Pig and Wannan Black Pig(P>0.05), showed the trend with BB>Bb>bb, and B allele have made the increase trend in IMF content.This study provides preliminary basis for H-FABP gene research, as a candidate gene, that influence carcass traits and meat quality.
