| According to their structure and functions, DNA methyltransferases(DNMTs) aredivided into two major families:maintenance methyltransferase (DNMTs) and denovo methyltransferases (DNMT 3a, DNMT3b), italso inclus DNMT2.To investigate the mRNA abundance ofporcine malic DNMTsgenesacross theadipose and muscle tissues at different body site of Landrace (a leaner Western breed) Rongchang pigs and Tibetan pigs(two fatty indigenous Chinese breeds), The real-time fluorescence quantitative PCR was applied. In addition, the relationships betwin on adipose deposition and muscle growth traits and gene expression changes are analysed.Our results showed that theexpression changes of porcineDNMTl, DNMT2, DNMT3a and DNMT3bmRNA in the adipose and muscle tissues exhibited the specific different patterns bygenderes and breeds:The mRNA abundunce of DNMT1 mRNA in female Rongchang pigs was significantly higher than in female Landrace pigs in Abdominal subcutaneous adipose (P<0.05), the mRNA abundunce of DNMT 1 mRNA in female Rongchang pigs was significantly higher than in female Landrace pigs in all tissues except Abdominal subcutaneous adipose (P<0.01). The mRNA abundunce of DNMT1 mRNA in female Rongchang pigs was significantly higher than in female Tibetan pigs in Upper layer of backfat, Inner layer of backfat, Great omentum, Retroperitoneal adipose, Intermuscular adipose and Pericardial adipose(P<0.01). The mRNA abundunce of DNMT1 mRNA in female Tibetan pigs was significantly higher than in female Landrace pigs in Great omentum, Retroperitoneal adipose, Intermuscular adipose, Longissimus dorsi muscle and Psoas major muscle(P<0.01). The mRNA abundunce of DNMT1 mRNA in male Landrace pigs was significantly higher than in male Rongchang pigs and male Tibetan pigs in all tissues except Inner layer of backfat (P< 0.01), The mRNA abundunce of DNMT1 mRNA in male Rongchang pigs was significantly higher than in male Tibetan pigs in Pericardial adipose, Lesser omentum (P<0.01).The mRNA abundunce of DNMT2 mRNA in female Landrace pigs was significantly higher than in female Rongchang pigs and female Tibetan pigs in Upper layer of backfat, Inner layer of backfat, Great omentum, Abdominal subcutaneous adipose, Intermuscular adipose, Pericardial adipose and Psoas major muscle (P< 0.01). The mRNA abundunce of DNMT2 mRNA inmale Landrace pigs was significantly higher than in male Rongchang pigs and male Tibetan pigs inlnner layer of backfat, Great omentum, Retroperitoneal adipose, Abdominal subcutaneous adipose, Intermuscular adipose, Pericardial adipose, Longissimus dorsi muscle and Psoas major muscle (P<0.01).But, The mRNA abundunce of DNMT2 mRNA inmale Rongchang pigs was significantly higher than in male Tibetan pigsinUpper layer of backfat and Inner layer of backfat(P<0.01).The mRNA abundunce of DNMT3a mRNA in female Landrace pigs was significantly higher than in female Rongchang pigs and female Tibetan pigs in all adipose tissues (P<0.01).But, The mRNA abundunce of DNMT3a mRNA in female Rongchang pigs was significantly higher than in female Landrace pigs and female Tibetan pigs inLongissimus dorsi muscle and Psoas major muscle(P<0.01). The mRNA abundunce of DNMT3a mRNA inmale Landrace pigs and male Rongchang pigs was significantly higher than male Tibetan pigs inRetroperitoneal adipose and Intermuscular adipose(P<0.01).The mRNA abundunce of DNMT3b mRNA in female Landrace pigs was significantly higher than in female Rongchang pigs and female Tibetan pigs inlnner layer of backfat, Great omentum, Lesser omentum and Abdominal subcutaneous adipose(P<0.01). The mRNA abundunce of DNMT3b mRNA in female Rongchang pigs was significantly higher than in female Tibetan pigs inlnner layer of backfat, Retroperitoneal adipose, Abdominal subcutaneous adipose, Pericardial adipose, Longissimus dorsi muscle and Psoas major muscle(P< 0.01). The mRNA abundunce of DNMT3b mRNA inmale Rongchang pigs was significantly higher than in male Landrace pigs and male Tibetan pigs inAbdominal subcutaneous adipose and Intermuscular adipose(P<0.01).The mRNA abundance of DNMT1 mRNA in male Landrace pigs was significantly higher than in female Landrace pigs in Abdominal subcutaneous adipose(P<0.05). The mRNA abundance of DNMT1 mRNA in male Landrace pigs was significantly higher than in female Landrace pigs in all tissues except Abdominal subcutaneous adipose(P<0.01). The mRNA abundance of DNMT1 mRNA in female Rongchang pigs was significantly higher than in male Rongchang pigs inIntermuscular adipose, Pericardial adipose and Longissimus dorsi muscle(P< 0.01),in Retroperitoneal adipose(P<0.05). The mRNA abundance of DNMT1 mRNA in female Tibetan pigs was significantly higher than in male Tibetan pigs inLesser omenrum, Abdominal subcutaneous adipose, Intermuscular adipose and Psoas major muscle(P<0.05).The mRNA abundance of DNMT2 mRNA in female Landrace pigs was significantly higher than in male Landrace pigs in Retroperitoneal adipose and backfat(P<0.05) (P<0.01). The mRNA abundance of DNMT2 mRNA in male Rongchang pigs was significantly higher than in female Rongchang pigs inUpper layer of backfat and Pericardial adipose(P<0.05), in Retroperitoneal adipose(P< 0.01); The mRNA abundance of DNMT2 mRNA in female Tibetan pigs was significantly higher than in male Tibetan pigs inlntermuscular adipose, Intermuscular adipose and Psoas major muscle(P<0.05).The mRNA abundance of DNMT3a mRNA in male Landrace pigs was significantly higher than in female Landrace pigs in Retroperitoneal adipose, Intermuscular adipose,Longissimus dorsi muscle and Psoas major muscle(P< 0.01), in Upper layer of backfat, Pericardial adipose(P< 0.05). The mRNA abundance of DNMT3a mRNA in male Rongchang pigs was significantly higher than in female Rongchang pigs inUpper layer of backfat, Retroperitoneal adipose, Abdominal subcutaneous adipose and Intermuscular adipose(P<0.01).The mRNA abundance of DNMT3b mRNA in female pigs was significantly higher than in male pigs in Upper layer of backfat, Great omentum, Abdominal subcutaneous adipose and Psoas major muscle(P<0.01). |
