A method was developed for the extraction and determination of oleanolic acid in Cyclocarya paliurus. The performances of oscillation extraction, soxhlet extraction and ultrasonic extraction were investigated. On the basis of oleanolic acid, the results obtained by HPLC indicated that ultrasonic extraction had the highest extraction efficiency. According to the orthogonal design, The optimum ultrasonic extractions were established as follows:First,80% ethanol solution, ultrasonic extraction three times, extracting time 40 minutes, ultrasonic power 400W, solid to liquid ratio 1:40. In the system of HPLC, a Kromasil C18 column (4.6 mm×250 mm,4μm) was used with a mixture of acetonitrile-water (80:20) as the mobile phase, gradient elution, the flow rate was 0.9mL/min, and the detection wavelength was at 210 nm. This method was quick, accurate and can be used for the determination of oleanolic acid of Cyclocarya paliurus. Second, the contents of oleanolic acid present a law of low -high-low-high in leaves, a law of low-high-low in stems and a law of high-low-high in roots. The contents of oleanolic in leaves of September is high to 0.35 mg/g,but in stems is only 0.03 mg/g.In different ages, the triennial Cyclocarya paliurus have more oleanolic acid.Third, the spectrophotometry was used to determine the contents of total saponins.The results showed that the contents of oleanolic acid and total saponins in different organs have basic similar change rule. The contents of total saponins is the highest in leaves of September. According to above results, the best time for collecting leaves is in September and the best part is in leaves.
