Objective:To investigate the expression of transforming growth factor-β1(TGF-β1) and Endoglin in placenta and myometrium, and soluble endoglin and TGF-β1 in pregnancy-induced hypertension disease (HDCP) patients. To explore the role of TGF-β1,Endoglin and sEng in Pathogenesis of HDCP patients.Material and Methods:1.Object of research:one hundred and ten pregnant women among our inpatie-nts were randomly collected. All the subjects were divided into two groups (1) pregnancy-induced hypertension disease group(ninety women with pregnanc-y induced hypertension disease:thirty women with gestational hypertension, th-irty women with mild pre-eclampsia and thirty women with severe pre-eclamp-sia);(2)control group (twenty normal pregnant women), age ranging from 25 to 31 years, gestational age ranging from 33 to 39 weeks. they were diagnostic standard (Obstetrics and Gynecology,the Seventh Edition).two groups were pr-egnancy and first delivery. All the patients had no complications of internal medicine and other complications of pregnancy, and the pregnancy termination of bothgroups was cesarean section.2. Experimental method: Collect five milliliters blood of pregnant women in each group, and anticoagulation with EDTA-Na. Three thousand round per mi-nute,15 min centrifugation. The supernatant was stored at -80℃refrigerator. Each specimen (2cm x 2cm x 2cm) was collected from central placenta of placental maternal surface after cesarean section and myometrial tissue(1cm×1cm×1cm)of placenta attachment surface was also collected. Rinsed with sal-ine to remove blood and preserved at -80℃. ELISA (enzyme-linked immunos-orbent assay) assayed sEng and TGF-131 concentrations in peripheral blood of case group and control group and statistic comparison was performed.Real-time fluorescence reverse transcription polymerase chain reaction (PCR) to detect the Endoglin and TGF-β1 mRNA expression in placenta and uterine tissues of pregnant women. Results:1. The peripheral blood level of sEng in control group, pregnancy-induced hypertension disease group, mild pre-eclampsia group and severe pre-eclampsia group was2.930±0.400ng/ml,3.430±0.630ng/ml,5.940±0.530ng/ml,8.230±1.040 ng/ml respectively. And the level of serum sEng in case group was higher than which of control group, the difference was statistically significant (F=318.26, P<0.05).Diffenrence between two groups was statistically significant (P<0.05).2. The peripheral blood level of TGF-β1 in control group, pregnancy-induced hypertension disease group, mild pre-eclampsia group and severe pre-eclampsia was 9.850±3.190pg/ml,11.210±2.320pg/ml,14.320±4.370pg/ml,15.370±4.930pg /ml, respect-ively. And the level of serum TGF-β1 in case group was higher than which of control group, the difference was statisticallysignificant (F=116.27,P<0.05).Diffenrence between two groups was statistically significant (P< 0.05).3. The 2-△△ct volume of placenta Endoglin in control group, pregnancy-induced hypertension disease group.mild pre-eclampsia group and severe pre-eclampsia was 1.0,1.27±0.58,1.54±0.41,1.83±0.35respectively.And the level of placenta Endoglin in case group was higher than which of control group, the difference was statistically significant( F=4.87,P<0.05). Diffenrence between two groups was statistically significant (P<0.05).4. The 2-△△ct volume of placenta TGF-β1 in control group, pregnancy-induced hypertension disease group, mild pre-eclampsia group and severe pre-eclampsia was 1.0,1.64±0.33,1.92±0.38,2.23±0.53 respectively. And the level of placenta TGF-β1 in case group was higher than which of control group, the difference was statistically significant( F=5.04,P<0.05).Diffenrence between two groups was statistically significant (P<0.05).5. The 2-△△ct volume of myometrium Endoglin in control group, pregnancy-induced hypertension disease group, mild pre-eclampsia group and severe pre-eclampsia group was 1.0,1.32±0.46,1.59±0.37,1.93±0.52 respectively. And the level of myometrium Endoglin in case group increased, the difference was statistically significant F=5.16,P<0.05). Diffenrence between two groups was statistically significant (P<0.05).6. The 2-△△ct volume of myometrium TGF-β1 in control group, pregnancy-indu-ced hypertension disease group, mild pre-eclampsia group and severe pre-eclampsia group was 1.0.1.71±0.45,1.91±0.51,2.37±0.46 respectively.And the level of myometrium TGF-β1 in case group was higher than which of co-ntrol group, the difference was statistically significant( F= 4.97,P<0.05). Diffen-rence between two groups was statistically significant (P<0.05).7. The correlation analysis of serum sEng and placenta, myometrium Endoglin: serum sEng and the placenta Endoglin levels were positively related:pregnancy-induced hypertension group r=0.61 (P<0.05);mild pre-eclampsia group r=0.64 (P<0.05); severe preeclampsia r=0.58 (P<0.05).Serum sEng and the myometrim Endoglin levels were positively related:pregnancy-induced hypertension group r=0.59 (P<0.05), mild pre-eclampsia group r=0.62 (P<0.05), severe preeclampsia group r=0.61 (P<0.05).8. The correlation analysis of serum TGF-β1 and placenta,myometrium TGF-β1: serum TGF-β1 and the placenta TGF-β1 levels were positively related:pregna-ncy-induced hypertension group r=0.65(P<0.05),mild pre-eclampsia group r=0.64 (P<0.05), severe pre-eclampsia group r=0.60 (P<0.05). Serum TGF-β1 and the myometrium TGF-β1 levels were positively related:pregnancy-induced hyperte-nsion group r=0.64 (P<0.05), mild pre-eclampsia group r=0.61 (P<0.05), severe preeclampsia group r=0.65 (P<0.05).9. The correlation analysis of TGF-β1 and Endoglin of placenta, myometrium: Placenta TGF-β1 and the placenta Endoglin levels were positively related: pregnancy-induced hypertension group r=0.57(P<0.05), mild pre-eclampsia group r=0.61 (P<0.05), severe pre-eclampsia group r=0.60 (P<0.05). Placenta TGF-β1 and the myometrium Endoglin levels were positively related:pregnancy-induced hypertension group r=0.59 (P<0.05), mild pre-eclampsia group r=0.62 (P<0.05), severe preeclampsia group r=0.61 (P<0.05).Conclusion:1.The level of placenta, myometrium Endoglin in pregnancy-induced hypertensi- on group,mild pre-eclampsia group and severe pre-eclampsia group was ordinal rise, and higher than which of control group.2.The level of placenta,myometrium TGF-β1 in pregnancy-induced hypertension group, mild pre-eclampsia group and severe pre-eclampsia group was ordinal rise, and higher than which of control group.3.The concentration of serum sEng in pregnancy-induced hypertension group, mild pre-eclampsia group and severe pre-eclampsia group was ordinal rise,and higher than which of control group, and had positive correlation with the level of sEng in placenta and myometrium; The concentration of serum TGF-β1 in pregnancy-induced hypertension group,mild pre-eclampsia group andsevere pre-eclampsia group was ordinal rise, and higher than which of control group, and had positive correlation with the level of TGF-β1 in placenta and myome-trium. the placenta and myometrium TGF-β1 levels were positively related in every group, and the placenta and myometrium Endoglin levels were positively related in every group.4.Endoglin, TGF-β1 and sEng may be related to the occurrence of HDCP, and serum sEng and TGF-β1 may be a effective monitoring incidence of HDCP.
