Ganoderma lucidum, as a Chinese traditional medicine, has been used as a medicine and food for a long history. Ganoderma spore oil, a sort of lipin extracted from Ganoderma spore (the reproductive organs of Ganoderma lucidum), is rich in Ganodenic acid, unsaturated fatty acid, Reishi polysaccharides and so forth. The oil is an aggregation of effective components in Ganoderma lucidum. The reports on immunoregulation and antioxidation of Ganoderma spore oil are few at present. Cyclophosphamide (CPA) is common antineoplastic agents which can cause immu-nodepression. In order to reduce clinically immunocomprornised side effects caused by Cyclophosphamide and to prevent the aging caused by redundancy of free radicals in vivo, the immune deficient mice and the D-galactose aging mice are established respectivly and studied the immunoregulation effect and antioxidation capacity of Ganoderma spore oil on the mice.In the paper, the immunity effects and the immunoregulation mechanisms of Ganoderma spore oil are comprehensively studied. The oxidizing enzyme activity and its mechanisms of Ganoderma spore oil are detected.The main results were as follows:(1) Ganoderma spore oil can enhance the humoral immune function, the mononuclear, the phagocyte gobble up capacity and the NK cytoactive of the immune deficient model mice. It can enhance the humoral immune function and NK cytoactive of the normals, with significant difference from those of the normal controls(P≤0.05). Ganoderma spore oil cannot significantly enhance the cellular immune function and the mononuclear phagocyte gobble up capacity of the normals, with unsignificant difference from those of controls (P>0.05). Thus, Ganoderma spore oil can enchance the immunoregulation functions of the immune deficient mice and the normal mice according to The functional theory evaluating procedure and test method of health foods.(2) Ganoderma spore oil can significantly enhance the content of TNF-a and IFN-y in serum of the immune deficient model mice, the serum concentration TNF-a of normal mice and the expression of IL-2, IL-10, IL-12 and IL-4 mRNA of both model mice and the normal mice's splenic organ and thymus gland, with significant difference from controls (P≤0.05). The serum concentration IFN-y of the normals is different from those of controls (P>0.05). Ganoderma spore oil can be able to express higher cytokineion to immunoregulate the immune deficient model mice.(3) The activity of serum SOD and CAT in the aging mice and the activity of serum SOD in the normals can be enhanced by Ganoderma spore oil, with significant difference from those of controls (P<0.05). The activity of serum CAT in the normal mice are no difference from those of the controls (P>0.05). The serum antioxidant enzyme activity of both the immune deficient model and the normal mice can be enhanced by Ganoderma spore oil.(4) The expression of P21mRNA gene of the splenic organ and thymus gland in the normals and the the agings can be significantly reduced by Ganoderma spore oil, with significant difference from those of controls (P≤0.05). The expressions of C3mRNA gene can be enchance in the splenic organ and thymus gland of the model mice and only in thymus gland of the normals, with significant difference from those of controls (P<0.05). The expression in the splenic organ of the normals can not significantly enhance, with unsignificant difference from those of the controls (P>0.05).The lower expression of P21mRNA and the higher expression of C3mRNA might be one of the anti-aging mechanisms of Ganoderma spore oil.
