Study On Quality Standard And Fingerprint Of Ganoderma Lucidum Spore Oil

ObjectiveGanoderma lucidum fungus is the dry fruit body of Ganoderma lucidum(Ley-ss.ex Fr)Karst,or Ganoderma sinense Zhao,Xu et Zhang,belonging to the family of Ganoderma genus,Polyporaceae.Ganoderma lucidum has been used by traditional Chinese medicine for thousands of years of culture,with high medicinal and health value.Ganoderma lucidum spore is the the fungus’ s reproductive cells ejected from the cap of G.lucidum after the fruit bodies becoming mature,with all the inherited active substances of Ganoderma lucidum.There are triterpenes,polysaccharides,fatty acid,amino acid and some other functional composition.Ganoderma lucidum spore have resistive tumor and improve immunity much better than Ganoderma lucidum,clinical application also becomes wider.Ganoderma lucidum spore oil is a mixture oil extracting from Ganoderma lucidum spore by supercritical carbon dioxide extractive technology.There are triglycerides,fatty acid,triterpenes,Sterol and some other functional composition in Ganoderma lucidum spore oil,which has broad physiological activities,signifiant effects of anti-tumor,adjusting immunity and protecting liver.In recent years,there are a large number products of Ganoderma lucidum spore oil and its preparation products pouring into the Chinese market,and becoming expensive health products.It is of important social and economical value.But there are not much reserches reports on quality standards of Ganoderma lucidum spore oil and there are a lot of adulterants can not distinguished.So it is necessary to study on quality standards,which could be used for evaluating quality of Ganoderma lucidum spore oil to protect the people safe and effective medication.On the basis of previous deep study on chemical component,We establish the quality control and fingerprint of Ganoderma lucidum spore oil.Methods1.study on quality standardsThin-layer chromatography was used to make a thin-layer qualitative identification for Ganoderma lucidum spore oil.Optimize the preparation of AgNO3-chromatoplate and the developing the solvent system and examine the factor of temperature and humidity.To establish a quantitative method for the determination of 1,2-dioleoyl-3-palmitoyl-Glycerol in the Ganoderma lucidum spore oil by HPLC-ELSD with the optimized chromatographic condition.2.Study on the fingerprintsThe fingerprints were established of 11 batches of Ganoderma lucidum spore oil by HPLC-ELSD,and were analysed with fingerprint parameters common peaks and similarity analysis.Then,the experimental data were analysed with cluster analysis and principal component analysis by SPSS software.3.draft the quality standarsAccording to the Chinese pharmacopoeia(2010),combining with the characteristic of the mixture oil,we would draft the quality control in morphological identification and thin-layer identification and content determination and so on.ResultsThis experiment were studied and evaluated comprehensively from both qualitative and quantitative aspects.10%AgNO3-chromatoplate was preparated by immersing method,with petroleum ether-eyhyl acetate(5:1).Results TLC spot develops clear,good resolution,reasonable Rf.HPLC-ELSD was performed on a Diamonsil C18(250mm×4.6mm,5μm)with acetonitrile-Isopropanol(51:49)as the mobile phase and a flow rate of 1 mL · min-1.The temperature of drift tube was 40℃,and the flow rate of gas was 2.4 L · min-1.The average content of 1,2-dioleoyl-3-palmitoyl-Glycerol was 26.8%.HPLC-ELSD fingerprints of Ganoderma lucidum spore oil consisted of 10 common peaks,identifying 5 peaks.The similarity of S1-S11 is between 0.915 to 1.ConclusionThe quality standard and HPLC-ELSD fingerprint were established,the method is simple,accurate and repeatable,which can provide an evaluation of the quality of Ganoderma lucidum spore oil.
