[Objective] To explore acupuncture on climacteric syndrome effects of antioxidant stress, our research chose the ovariectomized SD female rats as the experimental animal models of climacteric syndrome. The experiments use of modern means of detection to detect the changes of reduced glutathione (GSH), oxidized glutathione (GSSG) and the changes of GSH/GSSG ratio in the uterus and the pituitary gland of the groups. For the purpose of assessing the antioxidant effects by acupuncture on ovariectomized rat model, to explore the possible mechanisms of acupuncture antioxidant effects on climacteric syndrome, and to provide the experimental basis of treating and preventing female reproductive endocrine diseases by acupuncture from modern science.[Method] Randomly divided 40 female SD rats of 4 months old (180±20g) into 4 groups: Electro-acupuncture(EA) group, medical group, ovariectomized (OVX) group, sham operation group, n=10, and remove both side ovaries to establish climacteric model. EA group: electro-puncture at "Guanyuan" (frequency:3Hz, current intensity:1-3mA, voltage:1-3V, 30min); Medical group:subcutaneous injection of estrostilben (2mg/100g),10 days for one course of treatment. Continuous treatment of 2 courses,3 days interval. Analyse the weight index changes of rats and the uterine index changes after treatment, and detect the GSH, GSSG content and and calculated the GSH/GSSG ratio of the uterine tissue and the pituitary gland. Use the SPSS 16.0 statistical software for statistical analysis.[Results] 1. Comparing with the OVX group the body weight of the EA and the medical group were reduced, and the change was significant (P<0.05), EA group was not as significant as medical group; 2. The uterine weight index of EA group was higher than OVX group, the difference is significant (P<0.001), comparing to the sham group, the difference was still statistically significant (P<0.001); 3. Comparing with the OVX group, the uterine GSH content of EA group increased (P<0.05), GSSG content decreased (P<0.05), GSH/GSSG ratio was significantly increased (P<0.001); 4. Comparing to OVX group, the GSH content in the pituitary increased, GSSG content decreased (P<0.05), GSH/GSSG ratio has significant statistical difference from OVX group and sham group.[Conclusion] 1. Electro-acupuncture can reduce the increased weight of the ovariectomized rats as perimenopause syndrome model, and increased uterine weight (uterine index), so Electro-acupuncture can control weight and reduce perimenopausal changes after the uterine contraction.2. Electro-puncture Guanyuan on ovariectomized rats can significantly increase the reduced GSH content, while decrease the already elevated GSSG levels, and also can improve the relative GSH/GSSG ratio, which indicate that acupuncture can improve the antioxidant system, raise the level of antioxidant capacity, reduce the metabolite of the free radical, thereby enhancing the body's resistance to disease and response capabilities, which play can delay senility.3. Electro-acupuncture therapy can improve the aging signs of pituitary and uterus on ovariectomized rats, the effects are better than estrogen therapy which is more gentle, has less side effects, and it has a obvious adjusting advantage.
