[Objective] The study is focus on the inhibition effect of the water extract of Chinese medicine "Pingliu Keli" on human glioma glioma SHG-44, and explores the mechanisms of the effect in an experimental research. [Methods] MTT assay was performed to measure the effect on the proliferation of SHG-44 cells treated by different concentrations of "Pingliu Keli (PK)". The apoptosis of SGH-44 cells was detected by flow cytometry. Western-blot revealed the expression of Bax, Bcl-2, poly-ADP-ribose polymerase (PRAP), Caspase-3, Caspase-9 in SHG-44 cell. [Result] PK significantly inhibited the cell viability of SHG-44 cells in a dose-dependent manner and induced SHG-44 cells apoptosis. It is suggested that PK could upregulate the expression of proapoptosic protein Bax and downregulate the antiapoptotic protein Bcl-2, with a concomitant increase in the levels of cleaved caspase-3, cleaved caspase-9 and cleaved PARP. [Conclusion] PK could inhibit the growth of SHG-44 glioma cell and promote the apoptosis of glioma cell in vitro, In the process caspase-3 may act as a potential mediator.
